摘要
以长春花叶片诱导的愈伤组织为材料,悬浮培养在含0.5mg/L的6-BA,40mg/L的Vc 50ml以及0.5mg/的2,4-D的MS培养基中,初始接入量﹑研究转速﹑蔗糖浓度和额外添加激素条件对愈伤组织增殖的影响.结果显示,在含有50ml MS液体培养基中接入3g愈伤组织,震荡转速设定为为120rpm,蔗糖浓度为15g/L,以及额外添加0.5mg/L的NAA时,愈伤组织增殖最快.研究结果为建立高效的愈伤组织悬浮培养细胞系奠定了基础.
Using callus induced from Catharanthus roseus leaves as experimental materials,the effects of rotating rates,primarily cultured callus amounts,sucrose concentrations and additional plant hormones on the proliferation of suspension cultured callus were investigated.The results showed that the callus of C.roseus grew with optimal proliferous rate when cultured in liqudid MS medium with 0.5mg/L 6-BA,0.5mg/L 2,4-D and 40 mg/L Vc,when 3g callus inoculated in 50mL mediun,rotating at 120rpm,with 15g/L sucrose and additional 0.5mg/L NAA in medium.The results provide the base for estalishment of high efficient suspension cultured cell of C.roseus.
