摘要 目的探讨细胞增殖相关基因p16,PCNA和CyclinD1在尖锐湿疣中的表达及意义。方法分别采用免疫组织化学和RT-PCR法检测30例尖锐湿疣、寻常疣、跖疣中细胞增殖基因p16,PCNA和CyclinD1的表达情况,并与30例正常包皮组织进行对照。结果 p16蛋白在尖锐湿疣、寻常疣、跖疣和正常对照中的阳性表达率分别为:26.67%,16.67%,6.67%和0;p16 mRNA的阳性表达率分别为:23.33%,16.67%,6.67%和0。疣皮损p16蛋白表达评分和mRNA光密度高于正常对照,差异有统计学意义(P<0.05)。PCNA蛋白在各组的阳性表达率分别为:93.33%,86.67%,76.67%和70.00%;PCNA mRNA的阳性表达率分别为:90.00%,76.67%,70.00%和70.00%。疣皮损PCNA蛋白表达评分和mRNA光密度高于正常对照,差异有统计学意义(P<0.05)。CyclinD1蛋白和mRNA在各型疣和正常对照阳性表达率均为0。结论 p16和PCNA基因在尖锐湿疣、寻常疣和跖疣的发生发展中起一定作用,尤其是对尖锐湿疣的促增殖作用显著。 Objective To study the expression of p16, PCNA and CyclinDl in CA. Methods Immunohistochemistry was used to assess the expression levels of p16, CyclinD1 and PCNA proteins in 30 CA, 30 verruca vulgaris, and 30 verruca plantaris, and RT-PCR was used to detect the RNA expression levels. Results Positive rates of p16 protein in CA, verruca vulgaris, verruca plantaris and normal controls were 26.67% , 16.67% , 6.67% and 0, respectively; Positive rates of p16 mRNA in verruca vulgaris, verruca plantaris and normal controls were 23.33% , 16.67% , 6.67% and 0, respectively. The expression score of p16 protein and its mRNA levels in verruca were higher than in normal controls (P 〈 0.05 ). The positive rates of PCNA protein in verruca vulgaris, verruca plantaris and normal controls were 93.33% , 86.67% , 76.67% and 70.00%, respectively. The positive rates of PCNA mRNA in verruca vulgaris, verruca plantaris and normal controls were 90.00% , 76.67% , 70.00% and 70.00% , respectively. The expression score of PCNA protein and its PCNA mRNA levels in verruca were higher than in normal controls( P 〈 0.05 ). No cyclinD1 protein and cyclinD1 mRNA were detected in all tissues. Conclusion p16 gene and PCNA gene play certain role in the development of CA, verruca vulgaris and verruca plantaris, especially in CA.
