摘要
滇蔗茅是甘蔗野生近缘属植物资源,通过光周期诱导甘蔗属热带种与野生滇蔗茅花期相遇进行远缘杂交利用,获得了远缘杂交F1代创新种质材料。本研究首先采用常规压片法对远缘杂交F1代材料云南03-316染色体数目进行鉴定,结果 2n=54条;进一步的双色基因组荧光原位杂交(genomic in situ hybridization,GISH)分析结果表明,杂交F1代细胞中含有15条来自父本云南95-20的染色体及39条来自母本越南牛蔗的染色体,远缘杂交过程双亲本染色体以n+n的遗传配对方式进行传递。从F1代间期核荧光原位杂交结果可以看出,来自双亲本的染色质以分散的方式单独分布。研究结果可为F1代创新材料的后续利用提供细胞学参考价值。
The Erianthus rochii Ken$ is the wild relatives of sugarcane, distant hybridization was took between Saccharum o ffic/narum L. and E. rochii Keng while it' s flowering meet by the photoperiodic induction, and Ft generation was obtained. The chromosome number in Ft was identified with 2n = 54 by conventional pressure plate method. And the l^sult of the 8enomic in situ hybridization (GISH) showed that there were 15 E. roc/di chromosomes and 39 S. o fficinarum chromosomes in F! progeny (Yunnan03-316). Chromosomal constitution of Fl hybrids derived from sexual cro^s between S. o.fficinarum L. and E. recJdi was n + n. The GISH result on nuclear of Ft progeny showed the chrome- tins came from male and female was not intermixed and stayed at different regions. The results could provide a cytology re^erence for the fol- low-up use of the Ft generation materials.
