摘要
香叶醇-10羟化酶(geraniol 10-hydroxylase,G10H)是中药秦艽中龙胆苦苷等环烯醚萜苷类成分合成途径的限速酶。本研究克隆了秦艽G10H基因,并对其基因及编码蛋白序列的特征进行了生物信息学分析,结果显示:秦艽G10H基因包含一个完整的长1491bp的ORF框,编码496个氨基酸;GmG10H与长春花、川西樟牙菜等植物G10H蛋白具有很高的同源性(≥69%),无跨膜结构域、信号肽等结构,可能定位于细胞质中。定量PCR结果显示,GmG10H主要在秦艽的花和根中进行表达。
Geraniol-10 hydroxylase is the rate-limiting enzyme in the iridoid glycosides biosynthesis of Gentiana macrophylla. In this paper, a GIOH gene (GmG10H) was cloned from G. macrophylla and characterized using bio- informatics methods. The results showed that GmG10H contains a completed ORF with 1 491 bp, encoding 496 amino acids. Protein sequence of GmG10H is similar to G10H proteins from Catharanthus roseus and Swertia mussotii with high homology (≥ 69%). GmG1 OH is predicted to be localized in the cytoplasm and without the transmembrane domain and signal peptide. Quantitative PCR analysis showed that GmG10H has high expression levels in the roots and flowers in G. macrophylla.
