摘要
目的 :研究脱氢表雄酮 (DHEA)诱导SD幼年雌性大鼠多囊卵巢综合征 (PCOS)动物模型的卵巢形态学及性激素变化 ,同时探讨其与胰岛素抵抗方面的关系。方法 :以DHEA皮下注射2 3日龄SDⅡ级雌性大鼠 ,分别观察卵巢体重、光镜 (HE染色 )及透视电镜改变 ,测定糖耐量、血清胰岛素、E2、T、P、PRL、FSH、LH水平以及卵巢颗粒细胞在体外不同刺激状态下 (空白、FSH、IGF 1及FSH +IGF 1)培养液中E2的含量。结果 :卵巢重量实验组显著高于对照组 (P <0 .0 5)。实验组卵巢呈多囊样改变 ,有 70 % (14 2 0 )的卵巢未见黄体形成 ,而对照组仅占 15% (3 2 0 )。卵泡直径显微测量实验组闭锁卵泡平均直径明显大于对照组 (P <0 .0 5)。实验组血清T、E2和空腹血糖水平明显高于对照组 (P值分别为 <0 .0 0 1、<0 .0 5、<0 .0 5)。空腹及服糖后 2h血胰岛素水平非常显著高于对照组 (P <0 .0 0 1)。空腹血胰岛素与空腹血糖乘积的倒数 (1 FINS×FGC)非常显著高于对照组 (P <0 .0 0 1)。卵巢颗粒细胞体外培养 ,其基础E2分泌量实验组明显大于对照组 (P <0 .0 5) ,两组颗粒细胞分别按不同组别加入FSH、IGF 1及FSH +IGF 1后 ,其E2分泌量均非常显著大于空白组 (P <0 .0 0 1)。结论 :DHEA诱导SD幼年雌性大鼠PCOS动物模型卵巢病理改变及血内分泌?
Objective: To investigate the morphologic and hormonal changes in the ovarium of polycystic ovarian syndrome (PCOS) rat model induced by dehydroeplandrosterone(DHEA),and the relationship between PCOS and insulin resistance. Methods: 23 day old female SD rats were randomly divided into two groups. The rats in group 1 were injected daily (s.c) with DHEA for up to 20 days and the rats in group 2 injected with oil at the same time. Ovarian weight, serum insulin levels and sex hormon levels in rat blood of both groups were determined . Oral glucose tolerance tests,light microscopic and electronic microscopic examine were performed. The contents of estradiol granulosa cell in culture medium under the condition with or without FSH or/and insulin growth factor Ⅰ (IGF Ⅰ) were measured. Results : The ovarian weight in group 1 was higher than that in group 2( P <0.05). The ovaria in group 1 showed multiple follicular cysts ,and the ratio of follicular cysts to normal follicle was significantly increased .70%(14/20) of the ovaria in group 1 had no corpus luteum,compared to 15%(3/20) in group 2. The diameter of atretic follicles determined under microscopy in group 1 was greater than that in group 2( P <0.05).The serum testeosterone and etrasdiol concentration were significantly higher in group 1 than those in group 2 ( P <0.001 and P <0.05 respectively),so as the fasting serum glucose ( P <0.05)and fasting serum insulin ( P <0.001).The value of 1/FINS×FGC was significantly higher in group 1 than that in group 2 ( P <0.001).The basic estradiol levels were higher in the granulosa cell culture medium of group 1 than those in group 2 ( P <0.05), and the estradiol levels were significantly increased under the stimulation by FSH or /and IGF Ⅰ in two groups. Conclusion: the morphologic and hormonal changes of the ovarium in the dehydroeplandrosterone (DHEA) induced rat PCOS model were similar to those of the patients with PCOS,and the insulin resistance was observed. It was proved to be a ideal PCOS amimal model.
