摘要
采用乙醇/硫酸铵双水相体系萃取茶叶中茶多酚。通过单一因素实验优化了影响茶多酚萃取率的各因素,通过正交实验确定了各因素的影响程度:即粗提液的用量>硫酸铵的用量>pH>乙醇的用量>分相温度。实验最佳提取条件为,当硫酸铵的质量分数为0.2 g/mL,乙醇的体积分数为50%,粗提液的体积分数为20%,pH为6.8,分相温度为40℃时,茶多酚的萃取率达到97.92%,分配系数为22.17。对测定样品进行加标回收试验,其回收率为98.6%~104.55%。对样品溶液进行十次平行测定测得标准偏差为0.235%。粗提液中茶多酚的浓度为0.365μg/mL。从而建立了一种从茶叶中萃取茶多酚的新方法。
Used ethanol / ammonium sulfate aqueous two-phase to extract tea polyphenols.By single factor experiment to optimize the factors affecting,the impact of various factors was determined by orthogonal experiment: the crude extract dosage the amount of ammonium sulfate pH the quantity of ethanol phase separation temperature.The optimum extraction condition: the mass fraction of ammonium sulfate is 0.2 g/mL,the volume fraction of ethanol is 50 %,the volume fraction of the crude extract liquid is 20 %,pH=6.8,phase separation temperature is 40 ℃,the extraction of tea polyphenols rate reached 97.92 %,the distribution coefficient was 22.17.Determination of the samples were spiked recovery test,the recovery rate was 98.6 %~104.55 %.For ten parallel determination,standard deviation was 0.235 % for the sample solution.The concentration of crude extracts of tea polyphenols was 0.365 μg/mL.Thus a new method was establish to extract tea polyphenol from tea.
