摘要
目的:采用核磁共振(NMR)代谢组学技术研究脾气虚、脾阳虚模型大鼠血浆代谢组学特征,探求与该证候相关的生物学本质。方法:以多因素复合的方法建立脾气虚、脾阳虚大鼠模型,检测实验动物脾虚证相关指标,探讨该二证型的相关性及差异性;以1H NMR技术对大鼠进行血浆代谢组学检测,分别采用弛豫编辑(CPMG)和扩散编辑(LED)脉冲序列观察大鼠血浆中小分子代谢物和脂类代谢物,探求与证候相关的生物标志物。结果:脾气虚、脾阳虚模型组血清D-木糖浓度、血清肌酸磷酸激酶(CPK)、血清胃泌素(GAS)、血浆胃动素(MTL)等均出现异常,其中血浆MTL水平在两个模型组呈不同变化趋势;脾气虚、脾阳虚模型组大鼠的血浆呈现出不同于正常组的代谢图谱,通过PLS-DA分析,得到了一些与证候相关的潜在生物标志物。结论:脾气虚、脾阳虚模型大鼠1H NMR代谢谱发生改变,该二证型主要存在能量代谢、脂代谢和糖代谢等异常,代谢组学技术有可能成为揭示中医证候本质的有力手段。
Objective: To explore the essence of spleen-qi deficiency pattern (SQDP) and spleen-yang deficiency pattern (SYDP) with a metabonomic study based on NMR technique. Methods: Induce model rats with SQDP and SYDP by combination of multi-methods, and detect corresponding indexes related to spleen deficiency so as to explore the difference and relevance between these two patterns. Detect the metabonomic profiles of plasma in model rats with 1H NMR spectroscopy so as to explore biomarkers related to the patterns. Results: Serum xylose, serum creatine phosphokinase, serum gastrin and plasma motilin all turned abnormal in both groups of SYDP and SQDP. Furthermore, the change of plasma motilin in rates with SQDP was opposite to it in rates with SYDP. The metabonomic profiles of plasma in both groups became abnormal, and some biomarkers related to these patterns were found after partial least square discriminant analysis. Conclusion: We found that the metabonomic profiles of plasma in rats with SQDP and SYDP were both different from normal rats, and a preliminary conclusion could be made that these two patterns were accompanied with abnormality in energy metabolism, glycometabolism and lipometabolism. Moreover, we can see possibility to determine the essence of patterns of TCM with techniques based on metabonomics in this study.
