丹参地上部分抗脑缺血活性部位筛选 被引量:6

Screening anti-cerebral ischemia effective composition of aerial part of Salvia

摘要 目的:筛选丹参地上部分抗急性脑缺血活性部位及有效成分检测。方法:选用小鼠常压耐缺氧模型和小鼠急性不完全性脑缺血模型,观察丹参地上部分提取物及氯仿、乙酸乙酯、乙醚、正丁醇四种不同极性溶剂萃取部位对小鼠耐缺氧时间和脑缺血后的存活时间,及小鼠脑匀浆后SOD、MDA、NOS含量的变化;同时采用HPLC对有效部位正丁醇萃取物进行成分分析。结果:当给药剂量为3.2g生药/kg时,丹参地上部分提取物及其正丁醇部位可显著增加小鼠常压耐缺氧时间,延长小鼠全脑缺血后的存活时间,同时可升高SOD活性、降低NOS活性、减少MDA含量。HPLC检测正丁醇萃取部位中含有丹酚酸B及丹酚酸A。结论:丹参地上部分具有抗急性脑缺血作用,活性成分主要分布在正丁醇部位,有效成分检测含有丹酚酸B。 Objective:Screening effective composition of aerial part of Salvia against acute cerebral ischemia in mice and determining the active composition with HPLC.Methods: Models of hypoxia tolerant experiment under normal pressure on mice was evaluated anti-hypoxic activities of different polar fraction extract of aerial part of Salvia.Models of acute incomplete cerebral ischemia was produced by the occlusion of bilateral common carotid arteries of mice was used to evaluate the survival time of the mice on cerebral ischemia by administrating different polar fraction extract of aerial part of Salvia.After death of mice,the activities of SOD and contents of MDA and NOS of rat's cerebral tissue was detected.At the same time the n-butanol extract were determined with HPLC.Results: After treated by extract of aerial part of Salvia and nbutanol fraction by oral administration at a dosage of 3.2g per kg,the survival time of mice in the states of anoxia and ligaturing bilateral carotid arteries was prolonged significantly(P 0.01).The activity of the SOD was increased.The contents of MDA and NOS were obviously reduced(P 0.01).The n-butanol extract contain salvianolic acid B.Conclusion: Normal butanol extract of aerial part of Salvia was the major activity site of against acute cerebral ischemic.The n-butanol extract contain salvianolic acid B.
