蛇莓水提物的体外抗菌活性及抗菌机制 被引量:11

In vitroantibacterial activity and mechanism of the water extract from Duchesnea indica Focke

摘要 为研究蛇莓水提物的体外抗菌活性及其作用机制,采用牛津杯法和肉汤稀释法检测了蛇莓水提物对不同菌株的体外抗菌活性;利用光学显微镜和透射电镜观察了亚抑菌浓度(0.5MIC)作用下,蛇莓水提物对金黄色葡萄球菌ATCC25923和大肠杆菌ATCC25922形态和超微结构的影响。结果显示,蛇莓水提物(400mg/mL)对金黄色葡萄球菌等5种标准菌株和不同临床菌株均有显著的抗菌活性;蛇莓水提物对金黄色葡萄球菌ATCC25923,大肠杆菌ATCC25922,沙门菌C79-13,藤黄微球菌CMCC28001和铜绿假单胞菌ATCC9027的最低抑菌浓度(MIC)分别是1.56,25,6.25,3.13和25mg/mL,最低杀菌浓度值是MIC的2倍;在亚抑菌浓度(0.5MIC)作用12h后,金黄色葡萄球菌集聚成团,大肠杆菌呈长菌丝;药物作用后菌体超微结构发生了明显改变,金黄色葡萄球菌细胞质壁严重分离,胞质空腔化。大肠杆菌表面粗糙,质壁分离和胞质空腔化不及金黄色葡萄球菌严重。结果提示,蛇莓水提物对不同菌种有较强的抑菌活性,主要作用于细菌细胞壁或细胞膜发挥抗菌活性。 To study the in vitro antibacterial activity and mechanism of the water extract from Duchesnea indica,Oxford cup and broth dilution methods were used to detect the antibacterial activity of D. indica. Light microscope and transmission electron microscope were used to observe the morphology and ultrastructures of Staphylococcus aureus ATCC25923 and Escherichia coli ATCC25922, which were treated by 0.5 minimum inhibitory concentration(MIC) of D. indica extract. D. indica extract(400 mg/mL) had obvious antibacterial activity against 5 standard strains and different clinical isolates. D. indica extract exhibited the MIC of 1. 56, 25, 6. 25, 3. 13 and 25 mg/mL against S. aureus ATCC25923, E. coli ATCC25922, Salmonella C79-13, Micrococcus luteus CMCC28001 and Pseudomonas aeruginosa ATCC9027,respectively. The value of minimum bactericidal concentration was as much twice as that of MIC. After the strains were treated with the extract at 0.5 MIC for 12 h,conglomerate S. aureus and long hyphostroma of E. coli were observed in morphology. The changes of ultrastruetures, such as cell membrane separated from cell wall, cavity emerged in cytoplasm and the uneven surface of cell, were observed in the treated S. aureus and E. coli. In conclusion,the water extract of D. indica has potent antibacterial activity against bacterium,and action mechanism of D. indica may be related to induce the changes of cell wall and cell membrane.
