摘要
建立了高效液相色谱法同时测定精油中5种禁用性激素炔诺孕酮、醋酸甲地孕酮、醋酸氯地孕酮、醋酸甲羟孕酮、丙酸睾丸酮含量的方法。精油样品采用90%甲醇-水(体积比)溶液提取,正己烷除脂后,以Kromasil100-5C18柱(250mm×4.6mm,5μm)分离,乙腈-水(65∶35,体积比)为流动相,二极管阵列检测器以242nm和290nm为检测波长进行定性和定量分析。该方法对精油中5种性激素分离效果良好,在100~10000μg/L范围内呈良好线性关系,相关系数大于0.9990;五种性激素的检出限为0.550~5.040mg/kg,定量限为0.662~5.260mg/kg;除丙酸睾丸酮外,实际样品中加标回收率为88%~120%,相对标准偏差<4%。该方法操作简便,定值准确,能够满足精油中性激素的检测要求。
A comprehensive analytical method based on high performance liquid chromatography (HPLC) has been developed for the determination of 5 sexual hormones (norgestrel, megestrol-17-acetate, chloromadinone 17 acetate, medroxyprogester- one 17 acetate, testosterone propionate) in essential oil. The cosmetic samples were extracted by 90% methanol water. The target compounds were separated on an Kromasil 100-5C18 column(250 mm×4.6 mm,5 μm) using acetonitrile-water(65/35) as mobile phases. The detective wavelength was 242 and 290 nm. The linear plots were obtained between 100 and 10 000 μg/L with good correlation coefficients larger than 0.9990. The limits of detection (LODs) for this method were in the range of 0.550-5.040 mg/kg, the qualification of detection (QOD) were between 0.662 5.260 mg/kg. The recoveries were in the range of 90%-120% except testosterone propionate. The relative standard deviations (RSD) were 〈4%. The results indicated that the method is simple, rapid, accurate and suitable for the determination of 5 sexual hormones in essential oil.
