摘要
对细胞质分别来源于提莫菲维 ( T.timopheevii) ,粘果山羊草 ( Ae.kotschyi) ,偏凸山羊草 ( Ae.ventricosa)的 3种普通小麦的雄性不育系 ,相应保持系和恢复系及其杂种的 mt DNA用 1 2个线粒体基因探针进行了 RFL P分析 ,结果为 :( 1 ) T、K、V型不育系的 mt DNA在组织结构上存在显著差异 ;( 2 ) T、K、V不育系的 mt DNA与共同的保持系间显著不同 ,推测 mt D-NA与小麦 cms有关 ;( 3)在 ATP6/Hind ,ATP6/Bam HI,2 6S/Hind ,2 6S/Bam HI组合中 ,检测到 T、K型不育系与其相应 F1代之间出现了多态性 ,证明核恢复基因影响 mt DNA的结构 ;( 4 )把雄性不育和育性恢复作为一个整体来考虑 ,着重比较不育系和 F1代之间的差异 ,并对其差异与 cms的关系进行了分析。
T ,K and V type wheat cms lines,of which the cytoplasms derived from T.timopheevii,Ae.kotschyi and Ae.ventricosa ,respectively,were compared with their common maintainer,K and V type restorer,T type restorer,and their F 1 hybrids with mitochondrial DNA (mtDNA) RFLP analysis.The results showed that:(1) difference were found between not only the K ,V and T type cms lines,but also cms lines and their restorers,so mtDNA may be associated with cms in wheat;(2) polymorphism was found between T ,K type cms lines and their F 1 hybrids in ATP6/Hind Ⅲ,ATP6/BamHⅠ, 26S/Hind Ⅲ,26S/BamHⅠ combinations,which revealed that nuclear genes influence the structure of mtDNA of cms lines;(3) “sterility fertility restoration” is an ideal model system to study the mechanism of cms.Comparative on difference of mtDNAs between cms line and its F 1 hybrid was emphasized in our study.
