摘要
目的 观察老龄小鼠异丙酚麻醉后对学习记忆及遗忘的影响。方法 选择正常昆明小鼠进行被动逃避避暗实验 ,分别于训练前 15min腹腔注射异丙酚 (5 0mg/kg和 10 0mg/kg)和训练后30min、3h腹腔注射异丙酚 (10 0mg/kg和 2 0 0mg/kg) ,记录训练后 2 4h的逃避电击进入暗室的潜伏期及 5min内的错误次数 ;处死动物 ,用放射免疫的方法检测小鼠脑中胆碱乙酰化酶的活性。结果 正常小鼠训练后 2 4h易于学习逃避电击进入暗室 ,潜伏期明显延长 ,给予小鼠 5 0mg/kg和 10 0mg/kg异丙酚潜伏期为 2 16± 0 2 7和 2 0 8± 0 36 ,与正常组比较明显缩短 (P <0 0 5 ,P <0 0 1) ;于训练后 30min即刻给予异丙酚 2 0 0mg/kg可出现逆行性遗忘 ,但于训练后 3h给药则未出现。异丙酚 (2 0 0mg/kg和10 0mg/kg)小鼠脑中胆碱乙酰化酶的活性于给药后 3h仍明显低于正常。结论 异丙酚可介导老年小鼠的顺行性和逆行性遗忘 。
Objective To investigate the effects of propofol on learning and amnesia in the old miceMethods The normal Kunming mice were trained in a one-trial, step-through, light-dark passive avoidance paradigm Propofol (50 or 100 mg/kg) was administered intraperitoneally 15 min before training ,and propofol (100 or 200 mg/kg) separately 30 min and 6 h in the immediate post-training period The latency of escaping into a darkened chamber and the wrong number of entering darkened chamber were recorded Animals were tested for recall at 24th h post-training time The activity of acetyltransferase was determined with radiommunoassay in the brain 3h after administrationResults All control animals were liable to learn the task as judged by their increased latencies to enter the darkened chamber at 24th h post-training time After administration with propofol 50 mg/kg or 100 mg/kg, the latencies were reduced (246 and 208,repectively), compared with control (P<005 or 001) The induction of retrograde amnesia by propofol(200 mg/kg) was significant 30 min after training (P<001),but did not appeared 3 h after training The activity of acetyltransferase decreased in propofol(100 mg/kg) group and propofol (200 mg/kg) group at 24th h post-administration time, compared with that in control groupConclusions Propofol can induce anterograde and retrograde amnesia in the old mice, which is associated with the inhibition of activity of cholinergic nervous system
