食管癌组织PTCH1基因和蛋白表达临床意义的研究 被引量:3

摘要 目的:探讨Hedgehog信号通路中PTCH1基因及蛋白在食管癌、癌旁组织和正常食管组织中的表达及其临床意义。方法:采用实时荧光定量PCR技术检测40例食管癌、其相应的癌旁组织和10例正常食管组织中PTCH1mRNA的表达,并应用免疫组化技术检测PTCH1蛋白的表达,分析其与患者年龄、性别、肿瘤大小、分化程度和转移等临床病理因素的关系。结果:食管癌组织PTCH1mRNA的表达为4.75(2.56~9.09),高于癌旁组织的0.80(0.4~1.52)和正常组织的1.00(0.592~1.69),差异有统计学意义,P<0.05。食管癌组织PTCH1蛋白表达率为75.00%,高于癌旁组织的52.5%和正常组织的40.00%,差异有统计学意义,P=0.036。鳞癌与腺癌组织PTCH1mRNA表达比较,P=0.017;高中分化与低分化比较,P=0.002。鳞癌与腺癌组织PTCH1蛋白表达比较,P=0.011;高中分化与低分化比较,P=0.038。PTCH1mRNA和PTCH1蛋白表达与食管癌组织分化程度和组织类型有关。结论:PTCH1基因和蛋白在食管癌中高表达,表明其可能与食管癌的发生发展有关,其检测可能成为食管癌临床诊断的新标志或治疗的新靶点。 OBJECTIVE: To investigate the expression of PTCH1 gene and protein about Hedgehog signal in pa- tients with esophageal carcinoma,precancerous tissues and normal tissues. METHODS: Using the real-time PCR and im- munohistochemistry to detect the expression of PTCHlmRNA in 40 cases esophageal carcinoma, the relative tissues and 10 cases normal esophageal tissues, and analyze its relationship with age, gender, tumor size, degree of differentiation and lymphatic moving. RESULTS: The expression of PTCH1 gene in esophageal cancer was 4.75 (2.56-9.09), which was higher than that of precancerous tissues 0. 80(0. 4-1. 52) and normal tissues 1.00 (0. 592-1.69). The difference was statistically significant(P%0.05). The expression of PTCH1 protein in esophageal cancer was 75.00% ,which higher than the expression in precancerous and normal tissues. The difference was statistically significant(P=0. 036). The expression of PTCH1 gene and PTCH1 protein was concerned with degree of differentiation and pathological type. The expression of PTCHlmRNA in squamous cell was higher than the expression in adenocarcinoma (P = 0. 017); the expression of PTCHlmRNA in highly and moderately differentiated cancer was lower than the expression in poorly differentiated cancer (P=0. 002). The difference was statistically significant. The expression of PTCH1 protein in squamous cell was higher than the expression in adenocarcinoma (P = 0. 011) l the expression of PTCHlmRNA in highly and moderately differentia- ted cancer was lower than the expression in poorly differentiated cancer (P=0. 038). CONCLUSIONS: The expression of PTCH l mRNA and PTCH1 protein is high in esophageal tissues, which may be concerned with the development of esopha- geal cancer. So the detection of PTCH1 gene and protein may become a new clinical diagnosis marker in patients with e- sophageal cancer or as a new target in treatment of esophageal cancer.
