摘要
目的观察血红素氧合酶-1(HO-1)与心脏瓣膜病并心房颤动(房颤)病人心肌重构关系,探讨HO-1在房颤心肌重构发生中的作用。方法选取风湿性心脏瓣膜病二尖瓣狭窄需行换瓣手术病人60例,将其分为窦性心律组(n=21)及房颤组(n=39)。外科换瓣手术时取右心耳组织约200mg,通过组织学检查、逆转录聚合酶链反应和Western Blot技术分别检测两组心肌纤维化的程度及HO-1的表达水平。结果与窦性心律组相比,房颤组HO-1mRNA及蛋白表达均明显上调(t=-4.75、-2.44,P<0.01)。结论心脏瓣膜病并房颤病人心房组织心肌重构过程中,HO-1表达显著升高,考虑HO-1表达增加与心肌细胞纤维化后细胞发生应激反应有关。
Objective To study the relationship between heme oxygenates-1 (HO-1) and myocardial remodeling in pa- tients with valvular heart diseases (VHD) complicating atrial fibrillation (AF). Methods Sixty patients with rheumatic mitral stenosis who needed to have a valve replacements were selected, and divided into two groups as sinus rhythm (SR) group (n =21) and AF group (n =39). The tissue of right auricle about 200 mg was taken at surgery, the degree of myocardial fibrosis and the expression of HO-1 were detected through h(stologieal examination, RT-PCR technique and Western blot, respectively. Results Compared with the SR group, the expression of HO-1 mRNA and protein in AF group were significantly up-regulated (t =-4.75, -2.44;P d0.01). Conclusion In the process of myocardial remodeling of AF patients, the increase of HO-1 expression is con- sidered to be related to the stress reaction of fibrosis of cardiac muscle cells.
