大鼠供肝保存时TET对HCA液的优化作用及与UW液的保存效果比较 被引量:1

摘要 目的通过用不同的保存液低温灌洗保存大鼠供肝2 h的实验,比较汉防己甲素(TET)-高渗枸橼酸盐嘌呤(HCA)溶液与UW(University of Wisconsin)液对供肝的保护作用,并分析TET对HCA液的优化效果。方法 SD大鼠80只,随机平均分成4组:A组(TET乳酸林格液组)、B组(HCA液组)、C组(TET-HCA液组)、D组(UW液组)。经实验处理后再检测各组肝组织的高能磷酸化合物的含量及保存液中丙氨酸氨基转移酶(ALT)、乳酸脱氢酶(LDH)值。结果 A组与B组各指标比较无显著性差异(P>0.05);C组与B组各指标比较无显著性差异(P>0.05);C组与D组各指标比较具有显著性差异(P<0.05)。结论在低温灌洗保存供肝条件下TET对HCA液具有积极的优化作用;TET-HCA液低温灌洗并保存鼠肝2 h,对供肝的保护作用具有与UW液类同的效果。 Objective To analyse the optimization effect on HCA of TET by comparing the preservation validity of TET-HCA and UW solutions on donor liver for 2 hrs perfused with different solutions. Methods 80 SD rats were divided into 4 groups perfused by TET-Ringer(A) , TET-HCA (B) , HCA (C) and UW (D) solutions, respectively. ATP in hepatocytes and ALT, LDH in preserved so- lutions were monitored and pathological changes of the preserved liver were observed after experiment. Results No significant changes were observed between group A and B. Instead, there are significant changes between group B and C, so were in groups C and D. Con- clusion TET-HCA has optimization effect on HCA under the cold preservation on donor liver. During 2 hrs preservation period, similar effect was found between TET-HCA and UW solutions.
