摘要
目的:观察中药益肾助孕颗粒对围绝经期大鼠卵巢血管内皮生长因子(vascularendothelialgrowthfactor,VEGF)表达的影响。方法:通过自然老化法构建围绝经期大鼠模型,选择处于围绝经期的大鼠(12~14月龄)40只,随机分为老年大鼠对照组、益肾助孕颗粒高、中、低剂量组。每组10只,分别用生理盐水2mL灌胃、益肾助孕颗粒按等效剂量高剂量10g·kg-1·d-1,中剂量5g·kg-1·d-1,低剂量为2.5g·kg-1·d-1,灌胃0.2mL·g-1·d“灌胃4周;另取5月龄大鼠10只作为青年大鼠对照组,生理盐水2mL灌胃4周。免疫印迹法定量检测各组大鼠卵巢VEGF蛋白质的表达;RT·PCR检测各组大鼠卵巢VEGFmRNA的表达。结果:治疗前,与青年大鼠组VEGF蛋白、VEGFmRNA比较,老年对照组和研究组大鼠卵巢的VEGF蛋白、VEGFmRNA都表达明显高于青年组,差异显著,有统计学意义(P〈0.05),用药4周后,与老年对照组大鼠卵巢VEGF蛋白、VEGFmRNA表达比,研究组大鼠卵巢VEGF蛋白、VEGFmRNA表达明显下降,差异显著,有统计学意义(P(0.05),并存在剂量依赖性。结论:益肾助孕颗粒能够下调围绝经期大鼠卵巢VEGF蛋白、VEGFmRNA的表达,可能通过调节VEGF治疗围绝经期综合征。
Objective: To observe the effect of the kidney Zhuyun particles on the expression of ovarian vascular endothelial growth factor (VEGF) of perimenopausal rats. Method: Build the perimenopausal rat model by natural aging and choose 40 rats (12-14 months) in perimenopausal. Divided them randomLy into the control group. The the kidney Zhuyun particles high, medium, low dose group (n = 10) of aged rats. Gavage the four groups separately with 2 mL of normal saline and the kidney Zhuyun particles of equivalent dose of 10, 5, 2.5 g ~ kg-l.d-1 , O. 02 mL ~ g-i-d-1 orally for four weeks; for another 5-month-old rats of 10 as a control group of young rats, and gavage them with 4 mL normal saline for four weeks. Assay for quantification of the rats ovarian VEGF protein expression in Western blot; RT-PCR was used to detect the rat's ovarian VEGF mRNA expression. Result: Before treatment, compared with group of young rats, the expression of VEGF protein and VEGF mRNA of the elderly control group and study group rats was significantly higher than the youth group. A significant difference was statistically significant ( P 〈 0.05 ). Four weeks after the treatment, the aged control rat's ovarian VEGF protein, VEGF mRNA expression was more than the research group rat's ovarian VEGF protein, VEGF mRNA expression. A significantly difference was statistically significant (P 〈 0.05 ). And exist dose dependence. Conclusion: The kidney Zhuyun particles can down to the perimenopausal rat's ovary menopause VEGF protein, VEGF mRNA expression, possibly by regulating VEGF treatment of perimenopausal syndrome.
