摘要
采用GC-MS对白木香内生真菌菌株Botryosphaeria rhodina A13接种于离体白木香树枝后产生的化学成分进行分析,寻找沉香组分从而建立一种离体实验方法。将菌株B.rhodina A13接种于已表面消毒的离体白木香枝条中,将接种后的枝条放入已灭菌的、垫有湿润滤纸的培养皿中,在黑暗、27℃环境中放置20 d,长满菌丝的白木香枝条用溶剂浸提处理后进行GC-MS分析。结果表明,实验组(菌株A13+树枝)、沉香对照组能检测到萜类化合物,两者都含有大根香叶烷类倍半萜5,9-二甲基-2-(1-甲基亚乙基)-环癸醇,而树枝对照组和菌株A13对照组中未能检测到沉香组成成分。内生真菌B.rhodina A13能诱导离体白木香树枝产生沉香倍半萜组分5,9-二甲基-2-(1-甲基亚乙基)-环癸醇,本研究建立了一种离体实验方法,能用于真菌诱导白木香形成沉香组分的研究。
In order to establish an in vitro experimental method, chemical constituents of Aquilaria sinensis excised twig inoculated with endophytic fungi Botryosphaeria rhodina A13 were analyzed by GC-MS to explore chemical eonstituents of agarwood. B. rhodina A13 was inoculated into surface-sterilized exeised twigs of Aquilaria sinensis. Each twig was then placed on a moist, sterile filer paper in a sterile petri plate. All plates were ineubated for 20 d at 27 ℃ in the darkness. Twigs together with myeelia on the surface were extracted with organic solvents. The extracts were analyzed by GC-MS. Terpenoids,sueh as 5,9-dimethyl-2-[ 1-methylethyldene] -1-cyclodecanol, sesquiterpene with germacrane skeleton in agarwood,were deteeted in experimental group (excised twigs inoculated with strain A13 ) and control group (ethanol extract of agarwood). No chemical constituent of agarwood in control group ( excised twig not inoculated with strain A13, and strain A13 cultured on PDA medium without excised twig) was detected. The results showed that B. rhodina A13 can induce Aquilaria sinensis excised twigs to produce 5,9-dimethyl-2-[ 1-methylethyldene]-1-cyclodeeanol, sesquiter- pene of agarwood. The in vitro experimental method was established to investigate the effeet of fungi on agarwoed forma- tion in Aquilaria sinensis.
