摘要
目的 :研究在光照下姜黄素诱导胃癌细胞凋亡的效应 ,探讨其作为光动力学治疗肿瘤新型光敏剂的可能性。方法 :体外培养的胃癌MGC80 3细胞加入姜黄素后光照处理 ;以台盼蓝拒染法计数细胞存活率 ,Giem sa染色观察形态变化并计数及统计凋亡指数 ,DNA琼脂糖电泳观察DNA片段化 ,流式细胞术检测细胞周期变化。结果 :光敏化姜黄素处理后 ,细胞存活率随药物浓度及处理时间依赖性下降 ,并呈现典型的凋亡细胞形态 ;5 μmol/L姜黄素光照处理细胞 2 4小时 ,即呈现典型凋亡梯形条带 ;经 2 5 μmol/L及 5 μmol/L光敏化姜黄素处理 ,细胞周期主要阻滞于G2 /M期 ,提示细胞的G2 /M期阻滞与光敏化姜黄素诱导MGC 80 3细胞凋亡有关。结论 :光敏化姜黄素具有显著诱导MGC 80 3细胞凋亡的作用 ,可望进一步开发成光动力学治疗肿瘤的新型光敏剂。
Objective: To investigate the effects of photoactivated curcumin on induction of apoptosis in human gastric cancer cell line MGC 80 3 Methods: MGC 80 3 cell was treated with curcumin plus fluorescence Experiments of dye exclusion counting and Giemsa staining were conducted to determine the cell survival, morphology, and apoptotic index The analysis of DNA fragmentation and cell cycles were performed by DNA electrophoresis and flow cytometer respectively Results: Cell viability dropped down dependent on the curcumin concentration and treatment time Contrarily, the apoptotic index increased dependent on the curcumin concentration and the typical apoptotic morphology “ladder bands”could be seen on DNA electrophoresis in 5 μmol/L curcumin plus light treated cells By analysis with flow cytometry, cell cycles showed G 2/M arrest after treatment with 2 5 and 5 μmol/L curcumin plus light, suggesting an association between G 2/M arrest and MGC 80 3 cell apoptosis induced by photoactivated curcumin Conclusion: Photoactivated curcumin has the effect of inducing cell apoptosis in MGC 80 3 cells, suggests a potential application of photodynamic therapy for tumor
