摘要
目的:探讨苦豆子提取物槐定碱(SR)I调控B7-H1基因对人胃癌细胞多药耐药的调节机制。方法:人胃癌细胞SGC7901和多药耐药细胞SGC7901/DDP常规培养,苦豆子提取物槐定碱干预48h后,MTT法检测SGC7901/DDP细胞对化疗药敏感性的影响,流式细胞仪分析细胞凋亡情况,流式细胞术检测B7-H1蛋白表达情况。结果:人胃癌多药耐药细胞SGC7901/DDP对DDP的敏感性明显低于SGC7901敏感细胞(P<0.01);槐定碱干预SGC7901/DDP细胞48h后,耐药细胞增殖受到抑制,凋亡率增加,B7-H1蛋白表达下调(P<0.05)。结论:槐定碱通过降低B7-H1基因表达,抑制细胞增殖,诱导其凋亡,逆转SGC7901/DDP细胞株的多药耐药性。
Objective: To investigate the reversal effect and mechanism of sophoridine(SRI) on human gastric cancer multidrug resistance (MDR) in SGC7901/DDP ceils. Methods: Human gastric cancer cells SGC7901 and MDR SGC7901/DDP were cultured conventionally. After treatment with SRI for 48h, the sensitivity of SGC7901/DDP cells to chemotherapy drugs was evaluated by the MTT assay, the expression of B7-H1 was evaluated by Western blot, and the effects of SRI on apoptosis and cell cycle were evaluated by flow cytometry(FCM). Results: The sensitivity of DDP in SGC7901/DDP cells was much lower than that in SGC7901 cells(P 〈 0.01). After treatment with SRI for 48h, cell proliferation was inhibited, apoptosis rate was increased, and the expression of B7-H1 was down-regulated in human gastric cancer multidrug resistance SGC7901/DDP cells(P 〈 0.05). Conclusion: SRI can reverse MDR in human gastric cancer cells SGC7901/DDP. The possible mechanism is related with down-regulating B7-H1 expression, inhibiting cell proliferation and inducing cell-apoptosis.
