摘要
目的检测鼠类肉瘤滤过性毒菌(V-raf)致癌同源体B1(BRAF)在正常大肠组织、大肠增生性息肉、大肠腺瘤及大肠癌组织中的表达,探讨其是否可以作为大肠癌早期诊断的分子标志物。方法采用免疫组化染色(S-P法)检测50例大肠癌组织、30例大肠腺瘤组织、10例大肠增生性息肉组织、10例正常大肠组织中BRAF的表达。结果 BRAF在正常大肠、大肠增生性息肉、大肠腺瘤及大肠癌组织中的表达率分别为0、40.00%、56.67%和78.00%,与大肠癌组织比较差异均有统计学意义(P<0.05)。BRAF的表达与组织学分级有关(P<0.05),而与大肠癌患者的发病年龄、性别、民族、肿瘤大小、临床分期、血清CEA、血清CA-199、有无淋巴结转移及远处转移无关(P>0.05)。结论 BRAF在大肠癌组织中高表达,其表达与大肠癌组织分化程度有关,提示其与大肠癌的恶性程度相关。
Objective To detect the expression of V - raf murine sarcoma viral oncogene homolog B1 (BRAF) in normal color- ectal tissue, colorectal hyperplastic polyps, colorectal adenoma and colorectal cancer expression, and to explore whether it can be used as early diagnosis of colorectal cancer of molecular markers. Methods BRAF expression in 50 cases with colorectal cancer, 30 cases with colorectal adenomas, 10 cases with colorectal hyperplasic polyps and 10 cases with normal colon tissue were detected by immunohis- tochemical ( S - P method). Results BRAF expression rates were respectively 0, 40.00%, 56.67% and 78.00% in normal colorectal tissue, colorectal hyperplasic polyps, colorectal adenomas, colorectal cancer, respectively. And comparing with colorectal cancer, there was significantly differences ( P 〈 0.05 ). BRAF expression was closely related with the histological grade ( P 〈 0.05 ). BRAF expres- sion was not correlated with colorectal cancer patient age, sex, ethnicity, tumor size, clinical stage, serum CEA, serum CA- 199, lymph node metastasis and distant metastasis (P 〉 0.05). Conclusion BRAF is highly expressed in colorectal cancer. The expression is related with the histological grade in colorectal cancer. This suggests that BRAF may be related to the degree of malignancy of colorectal cancer.
