摘要
目的:研究镇脑宁胶囊对实验性偏头痛的镇痛作用及脑内单胺类神经递质的影响。方法:采用利血平负荷血凝块注射建立实验性偏头痛小鼠模型,利用热板法观察镇脑宁胶囊对偏头痛模型动物痛阈的影响;荧光分光光度法测定小鼠脑内单胺类神经递质去甲肾上腺素(NE)、多巴胺(DA)、5-羟色胺(5-HT)、5-羟吲哚乙酸(5-HIAA)含量。结果:利血平负荷血凝块可使小鼠痛阈降低,单胺类神经递质显著降低,成功诱导实验性偏头痛小鼠模型。镇脑宁胶囊200-800 mg·kg-1可不同程度改善模型动物偏头痛症状,其中800 mg·kg-1剂量可显著提高痛阈值27%,分别提高脑内NE、DA、5-HT和5-HIAA含量达34%、33%、20%和30%。结论:镇脑宁胶囊可以治疗利血平化伴局部脑血管痉挛所致小鼠偏头痛。
This study was aimed to evaluate the effect of Zhen-Nao-Ning capsules on experimental migraine model mice. The experimental migraine model mice were induced by injecting reserpine injection and blood clots. The pain threshold was determined by hot-plate method and monoamine neurotransmitters including norepinephrine (NE), dopamine (DA), 5-hydroxytryptamine (5-HT) and 5-hydroxy indole acetic (5-HIAA) which were measured by fluorescence spectrophotometry method. The symptom of pain among animal models can be relieved in different degrees by the treatment of Zhen-Nao-Ning capsule at the dosage of 200-800 mg/kg. The pain threshold can be increased 27% after treatment of Zhen-Nao-Ning capsule at the dosage of 800 mg/kg. And contents of NE, DA, 5-HT and 5-HIAA were increased 34%, 33%, 20%, and 30%, respectively. The re- suits indicated that Zhen-Nao-Ning capsule can ameliorate migraine via regulating monoamine neurotransmitters.
