摘要
目的探讨山慈姑醇提取液对胃癌SGC-7901细胞的抗肿瘤机制。方法用山慈姑提取液对SGC-7901细胞进行干预,分为空白组、山慈姑提取液0.75、1、1.5 mg/mL组,MTT检测,激光共聚焦显微镜观察山慈姑提取液对胃癌细胞凋亡情况,透射电镜观察4组肿瘤细胞的超微结构变化。结果 MTT显示1.5 mg/mL组抑制率为62.39±2.56,与0.75 mg/mL及1 mg/mL组比较,P<0.05,呈剂量依赖性;激光共聚焦显微镜观察胃癌细胞1.5 mg/ml组凋亡细胞荧光强度最强,细胞核呈圆形或椭圆形变;透射电镜观察1.5 mg/mL组最明显,胃癌细胞核异染色质边聚成半月状,线粒体缩小明显,电子密度增加。结论山慈姑提取液能诱导胃癌细胞的凋亡,具有抗肿瘤作用。
Objective To observe the effect of pseudobulbus cremastrae seu pleiones extract(PCSPE) on human gastric carcinoma SGC-7901 cells and explore its anti-tumor mechanism.Methods SGC-7901 cells were divided into blank group,0.75 mg·mL-1 PCSPE group,1.0 mg·mL-1 PCSPE group and 1.5 mg·mL-1 PCSPE group.The anti-tumor effects of four groups were explored by MTT assay,the gastric cancer cells apoptosis induced by PCSPE was observed by laser confocal microscopy,and the ultrastructure of tumor cells was observed by transmission electron microscope.Results MTT results showed that the inhibition ratio in 1.5 mg·mL-1 PCSPE group was 62.39±2.56,the inhibition effect was the strongest among the groups and the difference was significant in a dose-dependent manner compared with other groups(P0.05).Laser confocal microscopy results showed that the fluorescence intensity of apoptotic cells in 1.5 mg·mL-1 PCSPE group was the strongest and the cell nucleus were circular or oval.Transmission electron microscope results showed that the heterochromatin of gastric cancer cells was demilune,the mitochondria was significantly reduced and the electron density was increased.Conclusion PCSPE can induce the gastric cancer cells apoptosis,and has good effects of inhibiting the growth of SGC-7901 cells.
