摘要
目的:探讨血管内皮抑制素的不同给药方法对A549肺癌细胞、脐静脉血管内皮细胞及BALB/c裸鼠移植瘤生长的影响。方法:用不同浓度的血管内皮抑制素作用于A549细胞及脐静脉血管内皮细胞,用MTT法检测其对细胞增殖的影响;构建BALB/c裸鼠A549移植瘤模型,观察不同给药方法下肿瘤体积、重量的变化;应用免疫组化法检测肿瘤组织中CD31和CD34分子的表达差异。结果:在各组不同浓度的血管内皮抑制素作用下,A549细胞增殖能力并未随抑制素浓度的增加而降低,仅在浓度为1μg/ml时与对照组有差异(P<0.05);脐静脉血管内皮细胞增殖能力与对照组比较均无统计学意义(P>0.05)。在体内实验中,单独使用血管内皮抑制素及血管内皮抑制素联合顺铂治疗,各实验组肿瘤体积较对照组均有不同程度的缩小(P<0.05),联合用药疗效优于单用血管内皮抑制素;单独使用血管内皮抑制素时隔日给药肿瘤抑制率最高(50.74%),联合用药3周(血管内皮抑制素1次/日+顺铂1次/周)抑瘤效果最好(96.41%);免疫组织化学结果显示各实验组肿瘤组织中CD31和CD34分子的表达均下降(P<0.05)。结论:血管内皮抑制素在体外浓度为1μg/ml时对A549细胞生长有抑制作用,而对脐静脉内皮细胞生长没有明显抑制作用。血管内皮抑制素与顺铂联合使用较单独使用的抑瘤效果明显。血管内皮抑制素联合化疗时,每日连续给药抑瘤效果更好。
Objective：To observe the effect about different dilivery methods of endostatin on A549 cells,vascular endothelial cells and transplanted tumor of BALB / c nude mice.Methods：To treat A549 cells and endothelial cells with different concentrations of endostatin,detected the cell proliferation by MTT assay,build BALB / c nude mice model inoculated A549 cells,observed changes of tumor volume,weight,detected differential expression of CD31 and CD34 molecules in the tumor tissue by immunohistochemical method.Results：Under the effect of different concentrations of endostatin,value-added A549 cells were not reduced while the concentration of endostatin increased,the group of 1μg/ml was different from the control group（P〈0.05）.The proliferation ability of endostatin was not statistically significant differenct compared with control group（P 0.05）.In vivo experiments,the tumor volume of each experimental group reduced varying degrees compared with the control group in the treatment of separate endostatin and combination of endostatin and Cisplatin（P〈0.05）,the effect of combined therapy were more effective than the separate.Inhibition ratios of the tumor weight was highest（50.74%） when endostatin was alone administrated every other day,the effect of antitumor was best（96.41%） when combined therapy of three weeks（endostatin 1 times/day＋cisplatin 1 times/week）.The expression of CD31 and CD34 molecules in the tumor tissue decreased in each experimental group showed by the results of immunohistochemistry（P〈0.05）.Conclusion：The growth of solid tumor was related with angiogenesis of the tumor.In vitro,endostatin inhibites the growth of A549 cells in the concentration of 1μg/ml,human umbilical vein endothelial cell growth was not significantly inhibites.Antitumor effect of endostatin combined with cisplatin was better than the endostatin only.Antitumor effect of endostatin combined with chemotherapy may be better while endostatin daily continuous administration,these provids a reference for choice of the endostar scheme.
