摘要
目的观察6-羟多巴(6-hydroxydopamine,6-OHDA)损伤大鼠黑质后迷走神经背核(dorsal motor nucleus ofthe vagus,10N)中乙酰胆碱转移酶(choline acetyltransferase,ChAT)和酪氨酸羟化酶(tyrosine hydroxylase,TH)含量的变化。方法 16只SD大鼠根据随机数字表分为6-OHDA大鼠组和对照组。6-OHDA大鼠组双侧黑质(substantia nigra,SN)内注射6-OHDA,对照组双侧SN内注射等体积的生理盐水。6周后断头取脑,Nissl染色观察SN和10N中的Nissl小体,免疫荧光和免疫印迹检测SN内TH和10N内ChAT与TH的表达。结果与对照组比较,6-OHDA大鼠组SN内TH阳性细胞数量从(54±5)个减少至(13±2)个(P<0.05),蛋白表达水平从0.62±0.13降低至0.34±0.11(P<0.05),10N中ChAT阳性细胞数量从(37±3)个减少到(26±5)个(P<0.05),TH阳性细胞数量从(6±3)个增加到(13±2)个(P<0.05),背侧延髓内ChAT的蛋白表达水平从0.07±0.02下降至0.05±0.02(P<0.05),TH蛋白表达水平从0.08±0.05增高到0.13±0.04(P<0.05)。结论 6-OHDA大鼠10N中ChAT和TH表达变化可能与帕金森病患者的胃排空障碍存在一定的联系。
Objective To observe the expression of choline acetyltransferase(ChAT) and tyrosine hydroxylase(TH) in rat dorsal motor nucleus of vagus(10N) induced by substantia nigra lesion by 6-hydroxydopamine(6-OHDA).Methods According to the random number table,a total of 16 female SD rats were randomly divided into a 6-OHDA group and a normal control group.6-OHDA was injected into the bilateral substantia nigra(SN) of the rats in the 6-OHDA group,and the same volume of saline was injected into the same position of the rats in the normal control group.After six weeks,the rats were sacrificed and the brains were obtained.Nissl staining was used to detect the Nissl bodies in the SN and 10N,and immunofluorescent assay and Western blot analysis were used to detect TH expression in the SN and ChAT and TH expression in the 10N.Results Compared with the normal control group,the number of TH immunoreactive neurons in the SN of the 6-OHDA group decreased from 54±5 to 13±2(P0.05),and the protein level of TH decreased from 0.62±0.13 to 0.3±0.11(P0.05).The number of ChAT immunoreactive neurons in the 10N of the 6-OHDA group decreased from 37±3 to 26±5(P0.05),but the number of TH immunoreactive neurons increased from 6±3 to 13±2(P0.05).In the dorsal medulla,the level of ChAT protein decreased from 0.07±0.02 to 0.05±0.02(P0.05),and the level of TH protein increased from 0.08±0.05 to 0.13±0.04(P0.05).Conclusion The changes of ChAT and TH expression in 10N may be associated with gastric emptying dysfunction in patients with Parkinson's disease.
