摘要
目的 观察在不同外钙条件和激动剂作用下新生大鼠脑细胞内游离钙浓度的变化及酮替芬 (ketotifen ,Ket)的影响。方法 通过荧光指示剂Fura - 2 /AM负载细胞 ,测定 [Ca2 +]i 值。结果 胞外正常钙 1.3mmol/L时 ,胞内静息 [Ca2 +]i 为(136 .49± 7.36 )nmol/L(n =10 )。Ket(5 0 0 μmol/L)对不同外钙条件下胞内静息Ca2 +浓度无显著影响 (P >0 0 5 ) ,Ket(5 0~5 0 0 μmol/L)可以剂量依赖性抑制KCl和L -谷氨酸引起的 [Ca2 +]升高。
Objective To observe the effects of Ket on free intracellular Ca 2+ ,the level of isolated newborn rat brain cells in presence and absence of extracellular KCl and L-Glu [Ca 2+ ] i was measured with a RF-5000 spectrofluorophotometer by preloading the cells with calcium sensitive fluorescent indicator Fura-2/AM. Result Resting [Ca 2+ ] iwas (136.49±7.36) nmol/L( n =10) in the presense of Ca 2+ 1.3 mmol/L in Hank's solution. No significant effects of Ket (500 μmol/L) on resting brain [Ca 2+ ] i level were observed. When extracellular Ca 2+ was 1.3 mmol/L,Ket(10~500 μmol/L concentration-dependently inhibited KCl and L-Glu-induceded [Ca 2+ ] i elevation. Conclusion Ketotifen can inhibit the voltage dependence calcium channel and receptor coperated calcium channel.
