摘要
目的观察安肺益肾方对慢性阻塞性肺疾病(COPD)大鼠肺功能和肺组织损伤的干预作用。方法健康Wistar大鼠72只,采用随机数字表法分为安肺益肾方大、中、小剂量组,氨茶碱组、模型组、正常对照组,每组12只,除正常对照组外,其余各组大鼠采用熏香烟加气管注入内毒素法建立COPD大鼠模型。造模成功后,安肺益肾方大、中、小剂量组分别给予生药2.4、1.2、0.6g/kg;氨茶碱组给药15mg/kg;模型组和正常对照组灌胃等量0.5%羧甲基纤维素溶液;每天灌胃1次,连续4周。各组随机取8只大鼠,测定肺功能,包括第0.2s用力呼气量(FEV0.2)、第0.2s用力呼气量与用力肺活量比值(FEV0.2/FVC)及呼气阻力(Re)、吸气阻力(Ri);取大鼠右肺HE染色,观察各组大鼠肺组织病理变化。结果与正常对照组比较,模型组FEV0.2/FVC、FEV0.2显著降低,Re、Ri显著升高(P<0.01)。与模型组比较,安肺益肾方大、中、小剂量组、氨茶碱组均能显著提高FEV0.2/FVC,降低Re(P<0.01);安肺益肾方大剂量组提高FEV0.2(P<0.05);安肺益肾方中剂量组提高FEV0.2,降低Re(P<0.01);安肺益肾方中剂量组FEV0.2/FVC高于安肺益肾方大、小剂量组,FEV0.2高于小剂量组(P<0.05),Re、Ri低于安肺益肾方大、小剂量组。安肺益肾方能减轻COPD模型大鼠肺组织病理炎性浸润,减少肺大泡的形成。结论安肺益肾方能改善COPD模型大鼠的肺功能,改善模型大鼠肺组织损伤。
Objective To observe the effect of Anfei Yishen Formula Lung-Quieting Kidney-Boosting Formula on pulmonary function and tissue injury in chronic obstructive pulmonary disease （COPD） rats. Methods Seventy-two healthy Wistar rats were randomized into Formula large, medium and small-dose groups, aminophylline group, model group and normal control group, with 12 in each. The COPD models were induced by intratracheal instillation of lipopolysaccharide and passive smoking. The rats in Formula large, medium and smalbdose groups were intragastrieally administered with Anfei Yishen Formula （crude drug 2.4g/kg, 1.2g/kg and 0.6g/kg respectively）. The rats in aminophylline group were administered with 15mg/kg of amin- ophylline. The rats in model group and normal control group were administered with 0.5 % of sodium carboxymethyl cellulose. The intragastric administration lasted for four weeks. Eight rats in each group were randomly selected to detect the pulmonary function such as forced expiratory volume in 0.2s （FEV0.2）, FEV0. 2-to-forced vital capacity （FVC） ratio （FEV0. 2/FVC.,%）, expiratory resistance （Re） and inspiratory resistance （Ri）. The pathological changes in lung tissue of the rats were observed under HE staining. Results Comparing with the normal control group, FEV0.2 and FEV0.2/FVC significantly decreased and Re and Ri significantly increased in the model group （P〈0.01）. Comparing with the model group, FEV0. z/FVC significantly increased and Re significantly decreased in Formula large, medium and small-dose groups （P〈0. 01）. FEV0.2 significantly in- creased in Formula large group （P〈0.05）. FEV0.2 significantly increased and Re significantly decreased in Formula medium group （P〈0. 05 or P〈0.01）. FEV0.2/FVC was higher and Re and Ri were lower in the Formula middle-dose group than in large and small dose group and the level of FEV0. 2 was higher than that of the smalbdose group. Anfei Yishen Formula could relieve the pathological inflammatory infiltration and reduce the formation of bullae in COPD rat lung tissue. Conclusion AnfeiYishen Formula could improve the pulmonary function and relieve the pathological changes of lung tissue in COPD rats.
