摘要
目的研究131I-chTNT单药或联合外放疗对荷瘤肺癌模型肿瘤生长的影响,并观察其在小鼠体内的分布。方法建立C57小鼠的Lewis肺癌模型,随机分组进行试验。测量给药后不同时间肿瘤组织及感兴趣器官的放射性。观察荷瘤鼠的生长延迟效应。结果注射药物后1d、3d瘤内注射131I-chTNT联合外放疗肿瘤内的放射性最高,其次依次为131I-chTNT瘤内注射、131I-chTNT尾静脉注射联合外放疗、131I-chTNT尾静脉注射,四组均有统计意义上的差别(P〈0.0001)。肿瘤生长延迟效应试验中,外照射组,尾静脉注射131I-chTNT组,外照射联合尾静脉注射131I-chTNT组,瘤内注射131I-chTNT组,外照射联合瘤内注射131I-chTNT组的绝对延长时间分别为:9.1±1.92d,3.2±1.54d,13.1±2.054d,5.7±1.78d,15.1±2.59d。结论外照射联合±后可促进其在小鼠体内肿瘤区域的浓聚,而对正常组织无影响。联合应用±可提高外照射对荷瘤小鼠的放射效应,同时瘤内注射要优于尾静脉注射131I-chTNT。
Objective To investigate the biodistribution of lewis carcinoma of mice with 131I-chTNT combines with or without irradiation,and to study the effects on the turnor growth of mice.Method We established the lewis lung cancer model.When tumor size was big enough,the mice were randomly divided into different groups to receive different treatments.The biodistribution of 131I-chTNT was observed at various times after injection in tumor-bearing mice.The effect of treatment was assessed by tumor growth delay.Result The uptake of 131I-chTNT in tumor tissue was highest in the group which received intratumor injection of 131I-chTNT combined with radiotherapy, the second highest was the group which only received intratumor injection,the third is the group which received tail-intravenous of the same drug combined with radiotherapy, and the last is the group which only received tail-intravenous injection.The absolute growth delay for irradiation group,only tail-intavenous of 131I-chTNT group,tail-intravenous combined with radiotherapy, only intratumor injection,intratumor injection combined with radiotherapy were 9.1 ±1.92d,3.2 ±1.54d, 13.1 ±2.054d,5.7 ± 1.78d, 15.1 ±2.59d,respectively. Conclusion 131I-chTNT combined with irradiation can increase the uptake of 13tI-chTNT in tumor without side effects.131I-chTNT can enhance the radiotherapeutic effect for tumor-bearing mice.And the way of injection of intratumor was better than of tail-intravenous.
