摘要
目的观察消癥散结、扶正活络方药对转化生长因子β1(TGF-β1)诱导的人肾小球系膜细胞增殖及Ⅳ型胶原分泌的影响。方法采用TGF-β1诱导肾小球系膜细胞增殖和Ⅳ型胶原表达作为肾纤维化的细胞模型,实验分为5组:空白组,TGF-β1处理组,消癥散结、扶正活络方药高、中、低剂量(终浓度分别为50 mg/L、100 mg/L和200 mg/L)组。MTT法检测肾小球系膜细胞的增殖抑制率,ELISA法检测肾小球系膜细胞Ⅳ型胶原的表达。结果与空白对照组比较,TGF-β1处理组细胞经TGF-β1诱导后,HMCs增殖明显,细胞内ECM物质Ⅳ型胶原含量显著提高,差异均具有统计学意义。与TGF-β1处理组比较,消癥散结、扶正活络方药组可抑制TGF-β1诱导的HMCs的增殖及Ⅳ型胶原的表达,差异均具有统计学意义。结论消癥散结、扶正活络方药对TGF-β1诱导的HMCs的增殖及Ⅳ型胶原的表达具有抑制作用,抑制TGF-β1刺激HMCs分泌细胞外基质可能是其抗纤维化形成的机制之一。
Objective To observe the effect of Xiaozhengsanjie Fuzhenghuoluo prescription on TGFβ1 - induced proliferation and type Ⅳ collagen secretion in the human mesangial cells (HMCs). Methods The Renal fibrosis of cells model was treated with TGF- β1 by inducing proliferation and type Ⅳ collagen secretion in HMCs. The experiment was divided into 5 groups: blank, control,low, medium, and high (50 , 100 ,200 mg/L) Xiaozhengsanjie Fuzhenghuoluo prescription groups. The proliferation and inhibition rate of HMCs was examined by MTT method and enzyme - linked immunosorben assay was used to detect the ex- pression of type Ⅳ collagen in cultured HMCs. Results The proliferation and the expression of type Ⅳ collagen of HMCs treated with TGF - β1 was significant than blank group. Xiaozhengsanjie Fuzhenghuoluo prescription at high concentration ( 200 mg/L) significantly inhibited the proliferation of HMCs. The production of type Ⅳ collagen was down regulated expression in Xiaozheng- sanjie Fuzhenghuoluo prescription groups compared with the control group. Conclusion Xiaozhengsanjie Fuzhenghuoluo prescrip- tion can inhibit TGFβ1 induced proliferation and expression of type Ⅳ collagen in HMCs. One of anti fibrosis mechanisms may be correlated with downregulating expression of type Ⅳ collagen in HMCs.
