摘要
本实验建立了用反相HPLC 法同时测定人血清中苯妥英及卡马西平的方法。以苯乙酮为内标,采用Ultrasphere ODS 柱(5μm)(φ4.6×150mm),甲醇:水(50:50)为流动相流速0.8ml/min 紫外210nm 检测,血清处理用于乙腈沉淀蛋白,样品预处理过程快速简便,整个过程仅需20min 即可检出。苯妥英最低检测浓度为0.626μg/ml,相对回收率为99.66~102.40%线性范围为1.252~40.06μg/ml,相关系数r=0.9999:卡马西平最低检测浓度为0.5μg/ml,相对回收率为101.28~103.93%,线性范围为1.274~20.38μg/ml,相关系数r=0.9999。将本法应用于临床治疗药物监测获得满意效果。
A Simultaneous quantitative method was reported for the determination ofphenytoin and earbamazepin in human Plasma by reversed phase HPLC.Aceto-phenone was used as an internal Standard.The HPLC was fitted with an ultras-phere ODS Column(5μm)(φ4.6×150mm).The mobile phase was consisted of 50%CH_3OH and 50% H_2O.The flow Vate was 0.8ml/min.The detection was carried outat 210 nm.Acetonitrile was used for the sedimentation of protein.The metohd rapidand convenient,all procedure could be completed within 20 minThe detection Limit of phenytoin was 0.626 μg/ml in Plasma.The relative rec-overy was 99.66%to 102.40%.The method was Linear within the range of1.252to 40.06μg/ml,and the coefficient of correlation was 0.9999. The detection Limit of carbamazepinc was 0.5μ(?)ml.The relative recovery (?)as101.28 to 103.93%.The method was Linear within the range of 1.274 to 20.38μg/ml,and the coej ficient of correlation was 0.9999.A satisfactory result was obtained for clinical therapeutic drug monitoring.
