黄芪扶正汤抗小鼠Lewis肺癌机制的实验研究 被引量:22

摘要 目的探讨黄芪扶正汤在提高Lewis肺癌小鼠免疫力的最佳剂量时是否具有抑制HIF-1α、VEGF表达的作用,为黄芪扶正汤在临床上的进一步应用提供实验依据。方法建立肺癌小鼠动物模型,随机分为4组:模型组、中药组、顺铂组、联合治疗组,每组7只。皮下接种后第7天,同时开始给药,顺铂组2mg·Kg^(-1),腹腔注射,每2日一次,共给药8次;中药组每日灌服黄芪扶正汤溶液10mL·Kg^(-1)·d^(-1),共15天;联合治疗组两药用法同前;模型组给予等量的生理盐水灌胃和腹腔注射,给药第15天用颈椎脱臼法处死小鼠,取瘤体称重并计算抑瘤率,免疫组化法检测肿瘤组织中HIF-1α、VEGF表达。结果中药组瘤重较模型对照组减轻(P<0.05);联合治疗组的瘤重低于顺铂组(P<0.05);联合治疗组的抑瘤率高于顺铂组、中药组(P<0.05)。免疫组化结果显示各治疗组HIF-1α、VEGF表达均较模型对照组减低(P<0.05)。结论黄芪扶正汤可抑制Lewis肺癌小鼠肿瘤生长,其机制可能与抑制肿瘤组织HIF-1α、VEGF表达有关。 Objective The purpose of this study is to define whether HFD ( Huangqi Fuzbeng Decoction)has the ability to suppress the expression of HIF-lct and VEGF in the best effective dose of improving the immune function, to provide evidence for the clinical application of HFD. Method Lewis lung cancer bearing mice were established and then randomly assigned into the following four groups (n=7, each group): model group, Huangqi Fuzbeng Decection(HFD) group, Cisplatin(DDP) group, HFD combined with DDP group. DDP group was treated with DDP 2 mg.Kg-1intraperitoneally once every other day for 8 times; HFD group was treated with HFD 10 mL.Kg -l orally once daily for 15 days; HFD plus DDP group adopted the same administration of HFD and DDP; Control group was treated with the same amount of normal sodium orally and/or intraperitoneally. The mice were sacrificed on the 15th day, stripping out the tumors tissiues and weighting. The tumor inhibition rate was calcuated. We detected the expression level of HIF-la and VEGF with IHC. Results The weight of the tumors in the model group were heavier than those of the HFD group (P〈O.05). The weight of the tumors in DDP plus HFD group was lighter than those of other groups (P〈O.05), The tumor inhibition rate is the highest in HFD and DDP treated group (P〈0.05). The result of immunohistochemistry showed the expression of HIF-la and VEGF in treated groups was significantly decreased when compared with the control group (P〈O.05). Conclusion Huangqi Fuzheng Decoction could inhibit the growth of the tumor on Lewis lung cancer beating mice. The mechanism may be correlated with the inhibition of HIF- 1 a and VEGF.
