颅脑爆炸伤后兔垂体AQP4和ACTH表达的实验研究 被引量:2

摘要 目的探讨水通道蛋白4(aquaporin protein 4,AQP4)对兔颅脑爆炸伤后垂体-肾上腺皮质轴的影响。方法 80只新西兰大白兔随机分为实验组(n=70)及正常对照组(n=10)。实验组建立兔脑爆炸伤模型,正常对照组不实施引爆。伤后1 h、6 h、12 h、24 h、72 h、7 d、14 d测量2组动物垂体含水量,采用免疫组化检测垂体组织AQP4及ACTH的表达,采用免疫双标法检测AQP4和ACTH的分布及共存情况。结果实验组垂体含水量、AQP4及ACTH的表达在伤后1 h即升高,72 h达到高峰,7 d后表达开始下调,与正常对照组比较,差异均具有统计学意义(均P<0.05)。实验组垂体含水量与AQP4表达呈正相关(r=0.886,P<0.05),AQP4与ACTH表达随时间变化呈正相关(r=0.959,P<0.001)。免疫双标结果显示:ACTH细胞胞质中可见AQP4和ACTH的表达。结论兔颅脑爆炸伤后,AQP4有可能参与垂体ACTH分泌的调节。 Objective To investigate the effect of aquaporin protein 4(AQP4) on pituitary-adrenal axis after explosive brain injury in rabbits.Methods Eighty New Zealand rabbits were randomly divided into experimental group(n=70) and normal control group(n=10).The explosive brain injury model was established in experimental group and no explosive injury was performed in normal control group.The water content was weighed,and expressions of AQP4 and ACTH in the pituitary tissue were detected in two groups by immunohistochemistry 1,6,12,24,72 h and 7,14 d after injury.The distribution and coexistence of AQP4 and ACTH were examined by double immunohistochemistry at the same time.Results Compared with normal control group,water content,expressions of ACTH and AQP4 increased 1 h,reached peak 72 h,and then decreased 7 d after injury in the experimental group.There were significant differences in water content,expressions of ACTH and AQP4 in above time points between the experimental group and normal control group(all P 0.05).In experimental group,there was positive correlation between water content and the expression of AQP4(r = 0.886,P 0.05),so between the expressions of AQP4 and ACTH over time(r = 0.959,P 0.001).The expressions of AQP4 and ACTH were observed in cytoplasm of ACTH cells by double immunohistochemistry.Conclusions AQP4 may be involved in the regulation of ACTH secretion in the pituitary after explosive brain injury in rabbits.
