摘要
目的建立较理想的SD大鼠胎鼠海马神经细胞体外原代培养方法。方法孕18 d(E18)SD大鼠的胎鼠,采用胰酶消化和机械分离相结合的方法进行海马神经元的原代无血清培养。结果在体外培养条件下神经细胞结构特征明显化,并能形成典型的神经细胞网络。结论该培养技术是海马神经细胞体外培养的理想方法。
Objective To establish a better primary culture method for the primary culture for hipocampal neurons of SD fetal rats.Methods Trypsin digestion and mechanical dissociation were adopted to conduct monolayer primary culture.Results Under the culture condition in vitro,fetal rat neurons showed similar form properties to their counterparts in vivo and typical nerve fiber net was formed.Conclusion This technique is an ideal tool for culture in vitro for neurons of hippocampal tissue.
