摘要
目的研究动力素作为肝癌相关抗原其在肝癌细胞株中的表达情况,评价其作为分子肿瘤标记以及肿瘤免疫治疗特异性靶位的可能性。方法运用RT-PCR技术检测国内建株的肝癌细胞株BEL-7404 kinectin基因(D2剪接区)mR-NA的表达,并与肝癌组织、相应癌旁肝组织、正常肝组织比较。结果肝癌细胞株BEL-7404中kinectin基因(D2选择剪接区)呈强阳性表达,肝癌组织中也呈较强的阳性表达,癌旁肝组织则表达呈弱阳性。结论 kinectin基因可作为肝癌诊断的辅助分子标记,并具有作为肝癌免疫治疗特异性靶位的潜在价值。
Objective To investigate the mRNA expression of kinectin gene in hepatocellular carcinoma(HCC) cell line. Methods With reverse transcripase polymerase chain reaction(RT-PCR),the mRNA expression of kinectin gene in BEL-7404 HCC cell line,hepatocellular carcinoma tissue and corresponding paraneoplastic tissue was tested and compared with normal liver tissue. Results Kinectin gene mRNA was expressed in BEL-7404 cell line and HCC tissue,low expression in paraneoplastic hepatocellular tissue,but negative expression in normal liver tissue. Conclusion The tumor antigen of kinectin can be used as molecular marker for early diagnosis and possible target for immunotherapy of human HCC.
