摘要
目的探讨肿瘤坏死因子(TNF)-α基因多态性及与幽门螺杆菌感染的胃溃疡发生的相关性,血浆TNF-α含量与吸烟的相关性。方法采用聚合酶链反应-限制性片段长度多态性技术结合琼脂糖凝胶电泳的方法,检测幽门螺杆菌感染的胃溃疡组及幽门螺杆菌感染的健康组的TNF-α基因多态性,同时采用酶联免疫吸附试验比较胃溃疡组吸烟者与不吸烟者的血浆TNF-α含量。结果携带A/A基因型为幽门螺杆菌阳性的胃溃疡患者发病的独立危险因素(OR=3.67,P<0.05),同时发现胃溃疡组吸烟者与非吸烟者相比,外周血TNF-α明显增高(P<0.05)。结论 TNF-α启动子区-308位点多态性与幽门螺杆菌感染的胃溃疡可能相关。吸烟能引起血浆TNF-α含量增高。
Objective To investigate the relationship between polymorphism of the tumornecrosis factor-alpha (TNF-α) gene and the risk of Helicobacter. pylori-infected (H.pylori-infected) gastric ulcer, the correlation between smoking with quantity of TNF-α in blood plasma. Methods H.pylori-infected gastric ulcer and H.pylori-infected healthy adults were genotyped by polymerase chain reaction-restriction fragment length polymorphism for the -308G/A polymor- phism in TNF-α gene. Enzyme-labeled immunosorbent assay were used to measure the level of TNF-αin smoker-unit and non-smoker unit of H.pylori-infected gastric ulcer. Results People carrying A/A allele had increased risk for H.pyloriinfected gastric ulcer (OR =3.67, P〈0.05). Compared with non-smoker unit, the level of TNF-α in blood plasma were significantly high in smoker-unit of H.pylori-infected gastric ulcer (P〈0.05). Conclusion Gene polymorphism of the TNF-αhas relationship with the risk of H.pylori-infected gastric ulcer, and smoking can increase the quantity of TNF-α in blood plasma.
