摘要
观察吗啡条件性位置偏爱(CPP)大鼠纹状体中谷氨酸递质和忖甲基-D-天冬氨酸(№仍A)受体NR2B亚基的变化,从受体前和受体层面探讨通过谷氨酸能系统的阿片类精神依赖机制。方法:吗啡剂量递增连续皮下注射10d,建立大鼠吗啡CPP模型,生理盐水组和吗啡模型组各取10只断头处死,取脑纹状体采用谷氨酸试剂盒测定谷氨酸含量;另取6只经灌注固定后,免疫组织化学检测纹状体中NR2B的表达。结果:与生理盐水组比较,模型组大鼠纹状体谷氨酸含量和NR2B表达均增加。结论:纹状体中谷氨酸递质含量及其受体NR2B表达水平上调在一定程度上参与了在吗啡精神依赖的形成。
To explore the mechanism underlying the opiates psychical dependence by detecting the changes of glu- matic acid and NMI)A receptor 2B subunit (NR2B) in the striatum at the pre-receptor and receptor levels in conditioned place preference (CPP) rats. Methods: The CPP rat model was duplicated by subcutaneous injection of morphine for 10 days. The glumatic acid (Glu) and NR2B in the striatum were detected by colorimetry and irnmunohistochemistry respective- ly. Results: Compared with the rats injected with saline, the morphine dependent rats were exhibited with higher contents of Glu and NR2B. Conclusion: The up-regulated Glu and NR2B may contribute to the morphine-induced psychical dependence.
