四逆散对雷公藤多苷所致急性肝损伤的影响 被引量:23

摘要 目的研究四逆散对雷公藤多苷所致急性肝损伤的影响。方法将40只昆明种小鼠随机分为正常对照组、模型组、低剂量四逆散组、中剂量四逆散组和高剂量四逆散组,分别以相应剂量药物灌胃7 d,再以雷公藤多苷灌胃造模。观察各组的小鼠肝组织Bcl-2和Bax蛋白含量、Caspase-3的活性以及TNF-αmRNA的表达水平。结果模型组肝组织Bcl-2较正常对照组降低,Bax、Caspase-3、TNF-αmRNA较正常对照组升高。高剂量四逆散组Bcl-2较模型组升高,Bax、Caspase-3、TNF-αmRNA较模型组降低。低剂量和中剂量四逆散组较模型组相比,各项指标变化差异均无统计学意义。结论四逆散有对抗雷公藤多苷所引起急性肝细胞凋亡的作用。 Objective To study the effect of sini powder on acute liver injury induced by tripterygium glycosides.Methods 40 Kunming mice were randomized into a control group,a model group,a low-dose sini powder group,a middle-dose sini powder group and a high-dose sini powder group.The medicine gastric perfusion with corresponding doses was administered for 7 days.Additionally,the gastric perfusion with tripterygium glycosides was applied for modeling.The protein content of Bcl-2 and Bax,Caspase-3 activity and TNF-α mRNA expression in the liver tissue of mice were observed and compared among groups.Results In model group,Bcl-2 content was lower than that in normal control group,the results of Bax,Caspase-3 and TNF-α mRNA were increased as compared with normal control group.In high-dose sini powder group,Bcl-2 content was increased as compared with model group,the results of Bax,Caspase-3 and TNF-α mRNA were lower than those in model group.In comparison between low-dose and high-dose groups,there were no statistical differences in all the indexes.Conclusion Sini powder resists on acute hepatocellular apoptosis induced by tripterygium glycosides.
