摘要
采用荧光猝灭法以及紫外吸收差谱法研究了L-瓜氨酸与牛血清白蛋白在不同pH值下的相互作用机制,初步了解到L-瓜氨酸对牛血清白蛋白有猝灭作用,其猝灭机制为静态猝灭。利用Stern-Volmer方程求解出的各种参数表明pH值对两者的结合能力影响较大。同步荧光分析发现L-瓜氨酸的存在没有改变牛血清白蛋白的分子构象,依据Frster非辐射能量转移理论,确定了给体-受体间的结合距离和能量转移效率。
Fluorescence quenching and ultraviolet absorption subtraction have been employed to study the mechanism of interaction between L-citrulline and bovine serum albumin(BSA) at different pH.The preliminary results show that L-citrulline has the function of quenching BSA,and the quenching mechanism is static.Relevant parameters were obtained using the Stern-Volmer equation.The results showed that pH has a large impact on the binding ability of L-citrulline and BSA.Synchronous fluorescence analysis showed that the conformation of BSA was not changed in the presence of L-citrulline.The combination distance and the efficiency of energy transfer in the donor-acceptor pair were obtained using the non-radiative energy transfer theory of Frster.
