腺嘌呤致大鼠慢性肾功能衰竭模型研究 被引量:24

Research on Rat Model with Adenine-induced Chronic Renal Failure

摘要 目的通过不同剂量腺嘌呤所致慢性肾功能衰竭大鼠模型的比较,选出具备多种并发症的理想慢性肾衰模型。方法模型组大鼠分为M1、M2两组,M1以腺嘌呤150 mg/(kg.d)灌胃12周,M2以腺嘌呤300 mg/(kg.d)灌胃3周。观察大鼠一般情况并每周检测血压。分别于不同时间点检测血尿指标,并对肾组织进行光镜和电镜检查。结果 M1于实验第4周时血肌酐、尿素氮明显高于对照组,并呈进行性升高;第4周后分别于不同时间点陆续出现钙磷代谢紊乱、贫血、高血压等慢性肾衰并发症。M2血肌酐、尿素氮进行性升高,未出现贫血、高血压等并发症。两组大鼠肾脏病理学检查均以肾小管间质损害为主。M1于第4周时可见肾间质胶原纤维增多,纤维化指数明显高于对照组,并呈进行性增加。M2第3周时肾间质纤维化指数明显升高。结论腺嘌呤150 mg/(kg.d)灌胃12周可成功构建具备多种并发症的慢性肾衰大鼠模型,是慢性肾衰进展机制及其并发症防治研究的理想动物模型。 Objective To select the better one from the two rat models of chronic renal failure induced by different dose of adenine.Methods There were two model groups of rat:M1 and M2.M1 was given 12 weeks' adenine gastric gavage at a dose of 150 mg/(kg·d) while M2 given 3 weeks' adenine gastric gavage at a dose of 300 mg/(kg·d).The blood and urine were determined and all the kidneys were observed by light and electron microscope.Results In M1,there was an progressive increase of serum creatinine and blood urea nitrogen;after 4 weeks,the rats showed a series of complications consequently,such as anemia and hypertension.Rats of M2 showed an increase of serum creatinine and blood urea nitrogen but no anemia or hypertension.The characteristic of renal changes of both group was renal interstitial fibrosis.In M1,renal fibrosis was observed at 4 weeks,and it got more and more marked at 8 and 12 weeks.M2 showed marked renal fibrosis at 3 weeks.Conclusion 12 weeks' adenine gastric gavage at a dose of 150 mg/(kg·d) can develop an ideal rat model,which has a series of complications of chronic renal failure and is suitable to make research on pathogenesis and complications of chronic renal failure.
