制附子总碱的急性毒性及对过敏性鼻炎豚鼠鼻黏膜和组胺的影响 被引量:18

Study on acute toxicity of total alkaloids of processed Radix Aconiti Lateralis and its effects on blood histamine contents and pathomorphological changes in nasal mucosa in allergic rhinitis guinea pigs

摘要 目的:探讨制附子总碱对过敏性鼻炎(AR)豚鼠行为学指标、鼻黏膜病理形态和血液组胺含量的影响,同时测定制附子总碱的半数致死量(LD50),对其临床应用进行安全性评价。方法:40只FMMU豚鼠按体质量随机分为正常对照组、AR模型组、辛芩颗粒组、制附子总碱组,每组10只。采用卵清蛋白全身致敏与局部攻击方法制作AR豚鼠模型,正常对照组和AR模型组给予生理盐水,辛芩颗粒组每天给予辛芩颗粒4.65g/kg,制附子总碱组每天给予总碱提取物按生药量计1.13g/kg,观察豚鼠行为学指标和鼻黏膜病理形态的变化,测定血液组胺含量;对制附子总碱进行小鼠急性毒性实验。结果:制附子总碱组鼻痒、喷嚏、清涕等行为学指标及鼻黏膜炎症程度均有明显改善,血液组胺含量明显降低;制附子总碱的LD50为56.3g/kg制附子。结论:制附子总碱在临床用药方面是安全的;制附子总碱能有效缓解过敏性鼻炎的症状,降低血液组胺含量,改善鼻黏膜炎症的局部浸润,达到治疗过敏性鼻炎的目的。 Objective: To observe the effects of total alkaloids of processed Radix Aconiti Lateralis on blood histamine contents and pathomorphological changes in nasal mucosa of allergic rhinitis(AR) guinea pigs and assay LD50 of total alkaloids of processed Radix Aconiti Lateralis in guinea pigs to evaluate its safety of clinical application.Methods: 40 FMMU guinea pigs were randomly divided into control group,AR model group,Xinqin granule group and total alkaloids group,with 10 cases in each group.Generally sensitized guinea pigs were received repeated local sensitization in nasal cavity with ovalbumin.Guinea pigs in control group and model group were given normal saline,while guinea pigs in Xinqin granule group were given 4.65g/kg Xinqin granule and 1.13g/kg total alkaloids for total alkaloids group.Observe the pathomorphological changes in nasal mucosa and assay blood histamine contents in each group.Safety of clinical application of total alkaloids was proved through acute toxicity test on guinea pigs.Results: Total alkaloids of processed Radix Aconiti Lateralis can significantly reduce the amount of nasal secretions,sneezing and the degree of nasal scratching.The degrees of inflammation in nasal mucosal in total alkaloids group were improved and the contents of histamine were decreased obviously.LD50 of total alkaloids is 56.3g/kg.Conclusion: Total alkaloids of processed Radix Aconiti Lateralis were safe in clinical application.Total alkaloids of processed Radix Aconiti Lateralis play a significant role in the treatment of allergic rhinitis.
