摘要
目的研究白芍总苷(TGP)对大鼠胶原性关节炎(CIA)的作用及其可能机制。方法 SD大鼠随机分成6组:正常组、模型组、地塞米松(2 mg·kg-1)组及3个剂量(25,50,100 mg·kg-1)TGP组;用鸡Ⅱ型胶原建立胶原型大鼠模型;观察胶原型类风湿性关节炎(CIA)大鼠关节指数(AI)及关节组织的病理学变化;用ELISA法检测大鼠血清白介素-1β(IL-1β)和血管内皮生长因子(VEGF)的水平。结果白芍总苷可明显降低关节指数,抑制关节组织病理学改变;白芍总苷大、中剂量组能明显降低胶原性关节炎大鼠血清中IL-1β和VEGF的含量,与模型对照组比较,差异具有统计学意义(P<0.01)。结论白芍总苷对大鼠胶原性关节炎有明显抑制作用,其机制可能是与降低血清白介素-1β和血管内皮生长因子的水平、诱导炎性细胞因子网络趋于平衡有关。
Objective To research the total gluscosides of paeony(TGP) in the treatment of collagen-induced arthritis in rats and its possible mechanism.Methods SD rats were randomly divided into 6 groups: normal group,model group and dexamethasone(2 mg·kg-1) group,three dose TGP(25,50,100 mg·kg-1) groups.Type Ⅱcollagen-induced arthritis(CIA) model in SD rats was established.To observing the joint indexand the pathological changes of the CIA rats.The contents of interleukin-1β(IL-1β),vascular endothelial growth factor(VEGF)in CIA rats serum were measured with ELISA.Results TGP could inhibit the CIA rat joint index(P0.01),significantly reduced pathological changes.Total glucosides of paeonia can decrease the level of IL-1β,VEGF in CIA rats srums.The difference was significant compared with model group(P0.01).Conclusion TGP has an obvious inhibition on CIA rats and it′s mechanism maybe related to decreasing the contents of IL-1β,VEGF and to inducing the balance of the inflammatory cytokine network,and thus play a therapeutic role on CIA.
