摘要
目的分离纯化仙人掌多糖主要组分,观察仙人掌多糖主要组分(ODP-Ⅰa)对红细胞脂质过氧化损伤的影响,探讨其抗氧化机制。方法从野生仙人掌Opuntia dilleniiHaw.中采用超声提取法并用DEAE-Sepharose Fast Flow柱层析纯化得到仙人掌多糖组分ODP-Ⅰa、ODP-Ⅰb和ODP-Ⅱ′。以活性氧(H2O2)作用于离体正常大鼠红细胞,观察ODP-Ⅰa对红细胞溶血率、高铁血红蛋白含量、丙二醛含量、超氧化物歧化酶和谷胱甘肽过氧化物酶活性、红细胞膜蛋白巯基含量和膜蛋白质高分子聚合物的影响。结果 ODP-Ⅰa能够抑制H2O2诱导的红细胞溶血和高铁血红蛋白的产生;提高H2O2氧化损伤红细胞中超氧化物歧化酶活性、谷胱甘肽过氧化物酶活性和降低丙二醛含量;同时,能够抑制H2O2引起的膜蛋白巯基含量的降低和红细胞膜蛋白高分子聚合物的形成。结论 ODP-Ⅰa对活性氧引起的红细胞脂质过氧化损伤具有保护作用。
Objective To explore the effects of ODP-Ⅰa on erythrocyte lipid peroxidative injury,and its antioxidant mechanism.Methods ODP-Ⅰa,ODP-Ⅰb and ODP-Ⅱ′were obtained from Opuntia dillenii Haw.powder by ultrasonic extraction method and DEAE-Sepharose fast flow column chromatography.Normal erythrocytes of mice were treated with active oxygen(H2O2).The contents of ferrihemoglobin and malondialdehyde(MDA),the activities of Superoxide dismutase(SOD) and Glutathione peroxidase(GSH-Px) in red blood cells(RBC),the contents of the erythrocyte membrane protein-SH group and the production of high molecular aggregation(HMA) of erythrocyte membrane were detected.Results Compared with the control group,ODP-Ⅰa could suppress the RBC hemolysis and the production of ferrihemoglobin,and could also decrease MDA content and increase the activities of SOD and GSH-Px in erythrocyte.Moreover,ODP-Ⅰa could significantly inhibit the reduction of erythrocyte membrane protein-SH group content and the production of HMA of erythrocyte membrane.Conclusion ODP-Ⅰa has a protective effect against membrane lipid peroxidative damage induced by active oxygen.
