摘要
采用柠檬酸三钠还原法制备了胶体银用于标记羊抗人前白蛋白。在pH=7.0的Na2HPO4-NaH2PO4缓冲溶液中,银标记前白蛋白抗体与前白蛋白抗原产生特异性结合,在聚已二醇和氯化钾作用下,导致体系在470 nm处的共振散射强度降低。当前白蛋白浓度在0.0667~2.668μg/mL范围内,随着前白蛋白原浓度的增加,在470 nm处的共振散射强度线性增强,方法的检出限为0.0421μg/mL。,用于定量分析人血清中的前白蛋白,结果满意。
Colloidal silver was prepared by using trisodium citrate as reductive agent and was used to labele goat anti-human prealbumin.In the pH 7.0 Na2HPO4-NaH2PO4 buffer solution,the immune reaction between silver-labelled goat anti-human prealbumin and prealbumin took place.The labeled silver was released from the goat anti-human prealbumin and aggregated in the presence of Polyethylene glyco(PEG)-20000 and KCl,which leaded the intensity of resonance scattering peaks at 470 nm to drop greatly.The intensity of resonance scattering quenching is proportional to the concentrations in the range of 0.0667~2.668 μg/mL prealbumin.The detection limit is 0.0421 μg/mL,The assay was applied to quantitative determination of prealbumin in human serum,with satisfactory results.
