摘要
目的观察葛根素联合曲美他嗪对大鼠心肌缺血再灌注损伤的作用机制。方法将48只SD大鼠随机分为对照组、模型组、曲美他嗪组、葛根素联合曲美他嗪组(联合组),每组12只。对照组不结扎左冠状动脉前降支,模型组、曲美他嗪组、联合组结扎左冠状动脉前降支90 min,松解120 min制备急性心肌梗死再灌注模型,并于再灌注结束即刻,依次由尾静脉分别注射生理盐水、曲美他嗪、葛根素联合曲美他嗪,再灌注120 min后,取血检测心肌过氧化物酶、丙二醛和血清肌酸激酶水平,并观察心电图ST段抬高指数。结果联合组、曲美他嗪组血清肌酸激酶、过氧化物酶和丙二醛活性低于模型组(P<0.01),ST段抬高指数低于模型组(P<0.05);联合组血清肌酸激酶活性低于曲美他嗪组(P<0.05)。结论葛根素联合曲美他嗪可减轻SD大鼠心肌缺血再灌注损伤。
Objective To observe the mechanism of puerarin combined with trimetazidine on myocardial ischemia reperfusion injury in rats.Methods Forty-eight SD rats were randomly divided into control group,model group,trimetazidine group and puerarin + trimetazidine group with 12 rats in each group.Except control group,the other three groups were ligated left anterior descending artery for 90 minutes and were released for 120 minutes to establish acute myocardial infarction and reperfusion model.At the end of reperfusion,saline,puerarin and trimetazidine were respectively injected via the tail vein.The levels of peroxidase,malondialdehyde and serum creatine kinase were determined and ECG ST segment elevation indexes were detected in 120 minutes after myocardial ischemia reperfusion.Results The serum creatine kinase level,peroxidase and malondialdehyde activity,and ST-segment elevation indexes were significantly lower in trimetazidine group and puerarin + trimetazidine group than those in model group(P0.05,P0.01).The serum creatine kinase activity was lower in puerarin + trimetazidine group than that in trimetazidine group(P0.05).Conclusion The combination of puerarin and trimetazidine can relieve myocardial ischemia reperfusion injury in rats.
