Experimental Study on Apoptosis Effect of Histone Deacetylase Inhibitor SAHA Enhancing Cisplatin on Hepatocellular Carcinoma

摘要 目的:探讨去乙酰酶抑制剂SAHA与化疗药物顺铂(DDP)联合应用对肝癌细胞HepG2增殖的抑制效果。方法:通过SAHA联合顺铂处理培养的肝癌细胞HepG2,用倒置显微镜观察细胞形态学改变,通过MTT法检测细胞增殖抑制率,流式细胞术(FCM)检测细胞周期和凋亡率。结果:MTT结果显示,2.5μmol/L SAHA与6.25μg/ml DDP联合应用后5d,HepG2细胞增殖抑制率达(95.47±2.38)%,显著高于单用SAHA组的(49.36±1.73)%和DDP组的(42.52±1.35)%(均P<0.05);流式细胞术结果显示,SAHA与DDP联合应用明显导致细胞S期减少,G2/M期阻滞;SAHA联合DDP组细胞凋亡率为(16.26±1.29)%,显著高于单用SAHA组的(9.25±0.81)%和DDP组的(5.48±0.19)%(均P<0.05);倒置显微镜下可见,联合治疗组中细胞皱缩及细胞数量明显减少。结论:SAHA与DDP联合应用能显著提高对肝癌细胞HepG2增殖的抑制作用。 Objective:To investigate the inhibitory effect of suberoylanilide hydroxamic acid(SAHA) combined with cisplatin(DDP) on growth of human hepatocellular carcinoma cell line HepG2.Methods:Human hepatocellular carcinoma cell line HepG2 was treated with SAHA combined with DDP.Morphological changes of cell line HepG2 were observed by inverted light microscope.The growth inhibition rate of cells was analyzed by MTT assay,the cell cycle and apoptosis were detected by flow cytometry.Results:Five days after combined treatment with SAHA(2.5μmol/L) and DDP(6.25μg/ml),the growth inhibitory rate was(95.47±2.38)%,which was significantly higher than those in SAHA group(49.36±1.73)%(P<0.05) and DDP group(42.52±1.35)%(P<0.05).Flow cytometry displayed that combined administration of SAHA and DDP remarkably reduced S phase and arrested HepG2 cells at G2/M phase.The cell apoptotic rate was(16.26±1.29)% in SAHA and DDP group,which was significantly higher than those in SAHA group(9.25±0.81)%(P<0.05) and DDP group(5.48±0.19)%(P<0.05).After the treatment with DDP and SAHA,HepG2 cells showed some typical morphological features,such as cellular shrinkage and decrease in number of HepG2.Conclusion:SAHA combined with DDP may have an inhibitory effect on growth of human hepatocellular carcinoma cell line HepG2.
