远志茎愈伤组织无性系建立的研究 被引量:1

摘要 为保护野生资源,实现人工栽培,以远志的叶片、嫩茎和嫩根为材料,采用组织培养的方法,进行愈伤组织的诱导和分化,试管苗的生根、移栽和定植的研究,建立远志嫩根无性系。结果表明:MS+BA 0.5 mg/L+2,4-D 1.5 mg/L+NAA 0.5 mg/L是嫩根愈伤组织诱导培养和继代培养的理想培养基;MS+BA 0.8 mg/L+NAA 0.1 mg/L+AgNO3 1.8 mg/L和MS+BA 0.8 mg/L+NAA 0.1 mg/L+AgNO3 2.1 mg/L 2种培养基是嫩根愈伤组织分化培养的理想培养基;1/4 MS+NAA 0.2 mg/L是不定芽生根培养和试管苗生根继代培养的理想培养基;定植成活的试管苗保持了野生远志的所有生物学性状。 In order to protect wild resources and realize artificial cultivation, the leaves,tender stems and roots of Polygala Termuifolia were taken as material to investigate callus inducement and differentiation, rooting of tube seedlings and transplanting with the method of tissue culture, the clone of Polygcda tennuifolia from tender roots was established.The results indicated that the ideal medium for callus induction and multiplication of tender roots was MS+BA 0.5 mg/L+2,4-D 1.5 mg/L+NAA 0.5 mg/L;MS+BA 0.8 mg/L+NAA 0.1 mg/L+AgNO3 1.8 mg/L and MS+BA 0.8 mg/L+NAA 0.1 mg/L+ AgNO3 2.1 mg/L were both the best mediums for callus differentiation, 1/4 MS+NAA 0.2 mg/L was suitable for rooting of adventitious buds and tube seedlings.The test tube seedlings transplanted maintained all biological characteristics of the wild Polygala tennuifolia.
