响应曲面法优化微波辅助提取胭脂虫红色素工艺 被引量:14

Optimizing Conditions for Microwave-assisted Extraction of Cochineal Dye Using Response Surface Method

摘要 利用微波辅助水提取胭脂虫红色素,通过单因素试验考察了提取条件对色素得率的影响。结果表明,5 g胭脂虫干样,微波处理前常温浸泡40 min,液料比7∶1(g∶mL),微波功率700 W,辐射60 s后75℃水浴浸提40 min适宜于色素提取。在单因素试验的基础上,进行3因素3水平响应面分析试验,得到最优条件为5 g胭脂虫干样,液料比值7.5,微波功率714 W,辐射时间73 s,色素得率可达7.30%。最优条件下,合适的提取次数为12次,色素总得率达到29.95%,提取率为95.13%。 Cochineal(Dactylopius coccus Costa) dye was extracted by microwave-assisted extraction with water.Effects of single-factor test on the yield of cochineal dye was carried out.The results show that for 5 g cochineal,the suitable conditions for extraction of dye are cochineal sample soaked in water for 40min before microwave treatment,ratio of liquid to material 7 ∶1(g ∶mL),microwave power 700 W,microwave treatment time 60 s,and extract with water-bath at 75 ℃ for 40 min after microwave treatment.According to the single-factor test,the microwave-assisted extraction process parameters for extraction of cochineal dye were optimized using response surface method.The results show that the optimum parameters should be as follows: cochineal 5 g,ratio of liquid to material 7.5,microwave power 714 W,microwave treatment time 73 s.The extraction yield of conchineal dye is up to 7.30%.Under this conditon the reasonable extracting times is 12,the total yield of conchineal dye is 29.95 % and extraction yield is 95.13 %.
