β-榄香烯对肾癌786-0细胞凋亡的影响及其机制探讨 被引量:3

摘要 目的观察β-榄香烯对肾癌786-0细胞凋亡的影响,并探讨其可能机制。方法使用不同浓度β-榄香烯作用于786-0细胞24 h,MTT法检测细胞活力,流式细胞术检测细胞凋亡率,W estern b lot法检测细胞内磷酸化蛋白激酶B(p-Akt)及总Akt。结果随β-榄香烯浓度增大,细胞存活率逐渐降低,细胞凋亡率明显增加,细胞内p-Akt表达水平逐渐降低,均呈一定浓度依赖性(P均<0.05)。结论β-榄香烯可诱导肾癌细胞凋亡,其机制可能为抑制PI3K/Akt通路活性。 Objective To investigate the effect of β-elemene on apoptosis in human renal carcinoma cell 786-0 and the mechanism involved.Methods 786-0 cells were treated with different concentrations of β-elemene for 24 h,cell viability was measured by MTT assay,and apoptosis was evaluated by flow cytometry.Western blot was employed to detect the expression of protein kinase B(p-Akt) and total Akt.Results β-elemene inhibited the viability of 786-0 cells in a dose-dependent manner;it also induced apoptosis and inhibited the expression of p-Akt(all P0.05).Conclusion β-elemene can induce apoptosis in human renal carcinoma cell 786-0,inhibiting PI3K/Akt signaling pathway maybe the mechanism.
