摘要
目的建立HPLC法测定三花解毒饮中绿原酸的含量。方法采用Kromasil C18(4.6 mm×250 mm)色谱柱,以乙腈-0.5%磷酸溶液为流动相,进行梯度洗脱,柱温为35℃,紫外检测波长为326 nm,流速:1.0 ml.min-1。结果绿原酸进样量在0.162 3~0.814 9μg范围内与峰面积呈良好的线性关系,r=0.999 8(n=5),平均回收率为98.04%,RSD=0.97%(n=3)。结论此法专属性强,重现性高,可作为三花解毒饮含量控制标准。
Aim To assay the content of chlorogenic acid in Sanhua jiedu yin by HPLC.Methods With gradient elution,Kromasil C18(4.6 mm × 250 mm)was used as chromatograthic column with the acetonitrile 0.5% phosphoric acid as the mobile phase.The column temperature was 35 ℃,the ultraviolet detection wavelength for chlorogenic acid was 326 nm,and the flow rate was 1.0 ml·min-1.Results The progress of chlorogenic acid presented a good linearity with the peak square in the range from 0.162 3 to 0.814 9 μg,r=0.999 8(n=5).The average rate of recovery was 98.04%,and RSD was 0.97%(n=3).Conclusion This method is specific,reproducible and can be used as content control standards.
