天南星醇提取液诱导人K562细胞凋亡的实验研究 被引量:5

摘要 目的:探讨天南星醇提取物对人K562细胞的细胞凋亡作用。方法:采用四唑盐比色法(MTT)测定天南星醇提取物对人K562细胞的抑制作用,采用流式细胞术检测天南星醇提取液对人K562细胞凋亡的影响。结果:天南星醇提取液可明显抑制人K562细胞株的增殖,实验组与对照组间差异有统计学意义(P<0.01)。IC50达到65.07μg.mL-1。流式细胞学检查发现天南星提取物处理后的人K562细胞出现凋亡现象,且有一定的量效和时效关系。结论:天南星醇提取液能抑制体外培养人K562细胞的增殖,可能是通过诱导K562细胞的凋亡来实现的。 Objective: To investigate the apoptosis effects of alcoholic extract of rhizoma arisaematis on human K562 cells. Methods: The inhibition effect of alcoholic extract of rhizoma arisaematis on human K562 cell line was determined by MTT assay in vitro. The apoptosis induced by alcoholic extract of rhizoma arisaematis on human K562 cell line was detected by flow cytometry (FCM). Results: The alcoholic extract of rhizoma arisaematis significantly inhibited the proliferation of human K562 cells line. The difference between the experimental group and control group was statistical significance (P 〈 0.01). IC50 was 65.07 μg·mL^-1. The human K562 cell treated by rhizoma arisaematis showed apoptosis detected by FCM, which had certain quantity and time effect relationship. Conclusion: The alcoholic extract of rhizoma arisaematis obviously inhibited the proliferation of K562 cells in vitro, which might be associated with K562 cells apoptosis.
