摘要
目的:建立双向电泳和质谱技术探索维甲酸诱导甲状腺滤泡状癌凋亡的蛋白差异表达,为肿瘤治疗提供分子生物学证据。方法:无菌的含10%胎牛血清1640培养基培养甲状腺滤泡状癌细胞株FIE-133,分为正常对照组和实验组,实验组用维甲酸刺激24小时共同培养,提取对照组和实验组全蛋白,双向电泳分离蛋白质,采用PDQuest2-DE软件分析两组间差异表达的蛋白质斑点,用基质辅助激光解吸电离飞行时间串联质谱进行鉴定。结果:双向电泳图谱显示:实验组(维甲酸刺激组)与对照组的蛋白组存在显著性差异,质谱初步鉴定了5种主要蛋白。结论:双向电泳和质谱鉴定技术可以有效分离和分析维甲酸诱导甲状腺滤泡状癌FIE-133细胞凋亡的蛋白差异表达,为探索维甲酸治疗甲状腺滤泡状癌提供分子生物学的证据。
Objective:To identify main proteins in follicular thyroid neoplasm induced by retinoic acid.Methods: FIE-133 were suspended in RPMI 1640 and were cultured in the presence or absence of retinoic acid for 24h.The proteins were separated by 2-DE and then stained by silver staining.Using PDQuest software,high intensity proteins on the 2-D maps were selected and further analyzed by matrix-assisted laser desorption /ionization two-stage time of flight(MALDI-TOF) tandem mass spectrometry.Results: 5 differentially expressed proteins in 24h-experimental group and 24h-control group were found.Conclusion: The methods of 2-DE and MALDI-TOF-MS utilized in this study have effectively separated and identified the proteins in thyroid cancer induced by retinoic acid,and provides the evidence.
